全身低能量光療對重度燒傷大鼠早期全身炎癥反應(yīng)的影響

【摘要】:目的探討全身低能量光療(LLLT)對重度燒傷大鼠全身炎癥反應(yīng)的影響。方法將50只SD大鼠隨機分為空白對照組、單純燒傷組、燒傷后單次短時光照組、燒傷后單次長時光照組和燒傷后多次短時光照組,每組10只。單次短時光照組、燒傷后單次長時光照組、燒傷后多次短時光照組大鼠在燒傷后均給予低強度LED光(640nm)照射全身,分別照射5min/d、15min/d、5min×3次/d。各組大鼠分別于造模前1d,造模后即刻,造模后第1、3、7、14天取尾靜脈血檢測血清腫瘤壞死因子α(TNF-α)、白細胞介素(IL)-1β、IL-6的含量,計數(shù)靜脈血白細胞;并于造模后第3、7、14天測量創(chuàng)面面積,計算創(chuàng)面愈合率。結(jié)果與空白對照組相比,造模后第1天時單純燒傷組及燒傷后單次短時光照組大鼠血清TNF-α含量升高(P0.05),而燒傷后單次長時光照組及燒傷后多次短時光照組分別于造模后第1、7天時升高(P0.05);各組大鼠血清IL-1β含量均在造模后第1天時下降(P0.05),后逐漸恢復(fù),但恢復(fù)速度不一;單純燒傷組及燒傷后多次短時光照組大鼠血清IL-6含量均在造模后第1天時升高(P0.05),后下降,但單純燒傷組下降幅度大于燒傷后多次短時光照組,而燒傷后單次短時光照組于造模后第3天升高后下降,燒傷后單次長時光照組呈持續(xù)升高態(tài)勢(P0.05)。單純燒傷組及燒傷后單次長時光照組大鼠靜脈血白細胞計數(shù)均于造模后第1天時升高并維持(P0.05),其余各組白細胞計數(shù)無明顯改變。燒傷后單次短時光照組、燒傷后單次長時光照組和燒傷后多次短時光照組大鼠創(chuàng)面愈合率均高于單純燒傷組(P0.05)。結(jié)論全身LLLT可改善嚴(yán)重?zé)齻笫笱逖装Y因子TNF-α、IL-1β、IL-6的含量及靜脈血白細胞計數(shù),促進創(chuàng)面愈合,且不同照射模式所產(chǎn)生的效果不一。
[Abstract]:Objective to investigate the effect of systemic low energy phototherapy (LLLTT) on systemic inflammation in rats with severe burn. Methods Fifty Sprague-Dawley rats were randomly divided into blank control group, simple burn group, single light group after burn, single long time illumination group after burn and multiple short time light group after burn, with 10 rats in each group. The rats in the single short time illumination group, the single long time light group after burn and the burn multiple time short time illumination group were exposed to low intensity LED 640nm after burn. The rats were irradiated 5 min / d ~ 15 min / d ~ (-1) 5 min 脳 3 times / d ~ (-1) 路d ~ (-1) ~ (-1) 路min ~ (-1) ~ (-1) 路d ~ (-1) 路d ~ (-1) respectively. The blood samples of tail vein were collected at 1 day before model making and immediately after model making, and the contents of TNF- 偽 TNF- 偽 and IL-1- 尾 -IL-6 in serum were measured at 714 days after modeling, and the area of wound was measured on the 3rd day of 714 days after model making, and the contents of TNF- 偽 and IL-1- 尾 -IL-6 in serum were counted, and the wound area was measured at 714 days after modeling. The wound healing rate was calculated. Results compared with the blank control group, The content of TNF- 偽 in the serum of the rats in the burn group and the light group after burn was increased on the 1st day after the model, while the content of TNF- 偽 was increased in the single long time light group and the multiple short time light group on the 7th day after burn, and the level of serum TNF- 偽 in each group was higher than that in the control group on the 7th day after the model making, and the content of TNF- 偽 in the serum of the rats was increased in each group. The contents of IL-1 尾 in serum of rats decreased on the 1st day after the establishment of the model and gradually recovered. But the recovery rate was not the same, the serum IL-6 content in the burn group and the light group increased on the 1st day after the model, then decreased, but the amplitude of the decrease in the simple burn group was greater than that in the multiple light group after burn. However, the single short time illumination group increased at the 3rd day after burn, and then decreased, while the single long time light group showed a sustained increase trend of P0. 05% (P 0. 05%). The white blood cell count in vein blood of rats in burn group and single long time light group increased on the first day after burn injury and maintained P0.05A, but the other groups had no obvious change in white blood cell count. The healing rate of burn wounds in the group of single short time illumination after burn, the group of single long time light after burn and the group of multiple times light after burn were all higher than that of the group of simple burn (P 0.05). Conclusion systemic LLLT can improve the content of serum inflammatory factor TNF- 偽, IL-1 尾 and IL-6, and promote wound healing in rats with severe burn, and the effects of different irradiation modes are different.
