【摘要】：目的:探討頸內(nèi)動脈灌注冷生理鹽水誘導的選擇性腦部低溫對大鼠的安全性以及對急性腦缺血-再灌注損傷的治療效果。方法:60只SD大鼠隨機分為假手術(shù)組、正常灌注組、梗死組、局部低溫組、局部常溫組、靜脈低溫組。灌注前后監(jiān)測腦溫、肛溫以及各生理指標。再灌注24、48 h進行神經(jīng)行為學評分;再灌注48 h后檢測腦梗死體積及腦含水量。結(jié)果:頸內(nèi)動脈灌注15℃生理鹽水可使腦溫在10 min時到達33~34℃,停止灌注60 min后回升至灌注前水平。正常大鼠局部灌注冷生理鹽水后神經(jīng)行為學及組織學未見明顯異常,腦含水量與假手術(shù)組無顯著差異(P0.05)。灌注前后各個時間點生理指標差異無顯著性(P0.05)。局部低溫組其梗死體積、腦含水量、神經(jīng)行為學評分均顯著低于梗死組(P0.05)。而局部常溫組及靜脈低溫組各指標與梗死組比較差異均無統(tǒng)計學意義(P0.05)。結(jié)論:頸內(nèi)動脈灌注冷生理鹽水是一種高效、安全的選擇性低溫方法,可以顯著減少急性腦梗死的梗死體積及腦含水量,促進神經(jīng)功能恢復。
[Abstract]:Aim: to investigate the safety of selective cerebral hypothermia induced by cold saline infusion through internal carotid artery in rats and its therapeutic effect on acute cerebral ischemia-reperfusion injury. Methods Sixty Sprague-Dawley rats were randomly divided into sham operation group, normal perfusion group, infarction group, local hypothermia group, local normal temperature group and venous hypothermia group. Brain temperature, anal temperature and physiological indexes were monitored before and after perfusion. The neurobehavioral score was evaluated at 24 minutes after reperfusion and the volume of cerebral infarction and the water content of brain were measured after 48 hours of reperfusion.
