立體定向腦內(nèi)移植神經(jīng)干細(xì)胞改善顱腦損傷大鼠的神經(jīng)運動功能

【摘要】:背景:細(xì)胞替代治療是重建受損神經(jīng)系統(tǒng)組織結(jié)構(gòu),恢復(fù)神經(jīng)系統(tǒng)功能的一種有效策略,具有極為廣闊的應(yīng)用前景。目的:探討立體定向腦內(nèi)移植神經(jīng)干細(xì)胞對顱腦損傷大鼠神經(jīng)運動功能的影響。方法:將20只雄性SD大鼠隨機(jī)分為研究組和對照組,每組10只,應(yīng)用改良的自由落體方法制備腦損傷模型,傷后1 d研究組大鼠腦實質(zhì)內(nèi)移植胚胎神經(jīng)干細(xì)胞,對照組大鼠注射等量不含干細(xì)胞的培養(yǎng)液。在腦損傷前1 d、傷后1 d、1周、2周運用神經(jīng)學(xué)缺損評分法評價大鼠運動神經(jīng)功能。移植后2周,取腦組織分別行蘇木精-伊紅染色、抗BrdU、膠質(zhì)纖維酸性蛋白、β微管蛋白Ⅲ和酪氨酸羥化酶免疫組化染色。結(jié)果與結(jié)論:傷后1,2周研究組大鼠的神經(jīng)學(xué)缺損評分均顯著低于對照組(P0.05)。研究組大鼠受損腦組織中有較多的Brd U陽性神經(jīng)干細(xì)胞,一部分細(xì)胞呈膠質(zhì)纖維酸性蛋白、β微管蛋白Ⅲ和酪氨酸羥化酶陽性表達(dá),對照組大鼠受損腦組織中未見有Brd U陽性細(xì)胞。實驗結(jié)果表明,立體定向腦內(nèi)移植神經(jīng)干細(xì)胞可以在顱腦損傷灶中增殖分化,進(jìn)而顯著改善大鼠的神經(jīng)運動功能。
[Abstract]:Background: cell replacement therapy is an effective strategy to reconstruct the injured nervous system and restore the function of the nervous system. Aim: to investigate the effect of stereotactic neural stem cells transplantation on neural motor function in rats with craniocerebral injury. Methods: twenty male Sprague-Dawley rats were randomly divided into study group and control group with 10 rats in each group. The model of brain injury was established by modified free-fall method. Embryonic neural stem cells were transplanted into the brain of the study group 1 day after injury. Rats in the control group were injected with the same amount of culture medium without stem cells. The motor nerve function of rats was evaluated 1 day before and 1 week and 2 weeks after brain injury.
