硫酸鋅聯(lián)合阿司匹林對大鼠急性心肌梗死的保護(hù)作用及機制研究

【摘要】:目的:探討硫酸鋅聯(lián)合阿司匹林治療對大鼠急性心肌梗死的療效及其機制。方法:選取100只200 g左右的雌性SD大鼠,隨機分為5組,每組20只。假手術(shù)組不結(jié)扎并按常規(guī)飲食喂養(yǎng),其余4組結(jié)扎并在常規(guī)飲食的基礎(chǔ)上分別給以無菌蒸餾水(對照組)、硫酸鋅、阿司匹林、硫酸鋅聯(lián)合阿司匹林,連續(xù)給藥7 d后,使用超聲心動圖及侵入性導(dǎo)管檢測心功能及血流動力學(xué)指標(biāo),并對大鼠血液中的肌鈣蛋白T進(jìn)行檢測,然后取出心臟切片進(jìn)行HE和Masson染色檢驗,檢測各組心臟組織中的轉(zhuǎn)化生長因子-β1(transforming growth factor-β1,TGF-β1)、環(huán)氧合酶-2(cyclooxygenase-2,COX-2)、超氧化物歧化酶1(superoxide dismutase-1,SOD-1)和谷胱甘肽過氧化物酶-4(glutathione peroxidase-4,GPX-4)的m RNA和蛋白質(zhì)的表達(dá)情況。結(jié)果:給以阿司匹林和硫酸鋅聯(lián)合阿司匹林均能改善心肌梗死情況,保護(hù)受損心肌;與單獨使用阿司匹林比較,硫酸鋅聯(lián)合阿司匹林更能顯著提高收縮壓[(119±11)mm Hg vs.(103±7)mm Hg,P=0.037]、舒張壓[(98±4)mm Hg vs.(83±6)mm Hg,P=0.041]、平均動脈壓[(103±6)mm Hg vs.(89±5)mm Hg,P=0.029]及Emax[(5.27±0.33)mm Hg/μL vs.(4.13±0.29)mm Hg/μL,P=0.027];降低炎性侵潤、心肌梗死面積、心率[(328±11)beats/min vs.(364±19)beats/min,P=0.016]、全身血管阻力[(6.70±0.39)mm Hg×min/μL vs.(8.73±0.59)mm Hg×min/μL,P=0.039]及心肌梗死標(biāo)志物肌鈣蛋白T的水平[(583±51)pg/m L vs.(819±57)pg/m L,P=0.017];同時也更能降低促炎細(xì)胞因子TGF-β1和COX-2的m RNA[(0.65±0.18)vs.(1.48±0.12),P=0.014;(0.70±0.12)vs.(1.21±0.08),P=0.031]及蛋白[(2.42±0.19)vs.(3.61±0.27),P=0.032;(1.76±0.32)vs.(5.20±0.59),P=0.018)]表達(dá)水平,增加抗氧化酶SOD-1和GPX-4的m RNA[(2.14±0.11)vs.(1.58±0.12),P=0.021;(2.69±0.17)vs.(1.87±0.07),P=0.018]及蛋白[(5.40±0.62)vs.(2.40±0.33),P=0.036;(4.26±0.47)vs.(2.72±0.51),P=0.023]表達(dá)水平。結(jié)論:硫酸鋅聯(lián)合阿司匹林可有效保護(hù)急性心肌梗死對心肌的損傷,其主要的作用機制可能是通過抑制促炎細(xì)胞因子TGF-β1和COX-2,激活抗氧化酶類SOD-1和GPX-4的水平,實現(xiàn)抗炎抗氧化的作用,從而達(dá)到對急性心肌梗死的保護(hù)效果。
[Abstract]:Objective: to investigate the effect and mechanism of zinc sulfate combined with aspirin on acute myocardial infarction in rats. Methods: 100 female SD rats of about 200 g were randomly divided into 5 groups with 20 rats in each group. The sham-operation group was not ligated and fed on a conventional diet, while the other four groups were given aseptic distilled water (control group, zinc sulfate, aspirin, zinc sulfate combined with aspirin for 7 days). Cardiac function and hemodynamics were measured by echocardiography and invasive catheters. Cardiac troponin T was detected in the blood of rats. The heart sections were taken out for HE and Masson staining. The expression of m RNA and protein of transforming growth factor 尾 1(transforming growth factor- 尾 1 (TGF- 尾 1), cyclooxygenase-2cyclooxygenase-2 (COX-2), superoxide dismutase-1 (SOD-1) and glutathione peroxidase-4GPX-4 (Glutathione peroxidase 4GPX-4) in heart tissues were detected. Results: aspirin and zinc sulfate combined with aspirin could improve myocardial infarction and protect damaged myocardium, compared with aspirin alone.
