香煙煙霧對大鼠創(chuàng)面愈合的影響

【摘要】:目的 觀察香煙煙霧對大鼠創(chuàng)面愈合時間的影響;及對各組固定時間創(chuàng)面愈合率的影響;CD68單抗標(biāo)記免疫組化法計數(shù)各組創(chuàng)面愈合過程中巨噬細(xì)胞數(shù)量,探討巨噬細(xì)胞對創(chuàng)面愈合的可能影響。 方法 選取健康成年SD大鼠40只,體重190-260g,隨機分為4組:對照組(C組)、低焦油含量組(L組)、中焦油含量組(M組)和高焦油含量組(H組),每組10只。每組各只大鼠背部皮膚脫毛后畫出2cm直徑的4個圓圈,按畫定的圓圈剪掉皮膚全層,形成4個等大的圓形創(chuàng)面。各創(chuàng)面間距大于2cm。C組不做任何干預(yù),L組、M組、H組分別將大鼠置于自制的熏煙箱(30L)給予O.lmg、0.5mg、0.8mg不同等級焦油含量的香煙進行煙熏,每天30分鐘,直到創(chuàng)面愈合。觀察創(chuàng)面愈合時間,并于第7天及16天觀察創(chuàng)面愈合率各組分別與創(chuàng)面愈合的第3天、第7天取創(chuàng)面周圍組織,通過免疫組化法,單抗標(biāo)記CD68+,統(tǒng)計巨噬細(xì)胞數(shù)量[1]。 結(jié)果 創(chuàng)面愈合時間:高(H組)、中(M)組、低(L組)劑量組愈合時間明顯長于對照組(C),具有統(tǒng)計學(xué)意義(P0.05)。提示焦油含量高、中、低濃度組均有延長創(chuàng)面愈合時間的作用。高、中焦油含量濃度組較低焦油含量組更能延長愈合時間。 創(chuàng)面愈合率:與對照組(C組)相比,高(H組)、中(M組)、低(L組)焦油含量組第7、16天創(chuàng)面愈合率均低于對照組組,具有統(tǒng)計學(xué)意義(P0.05),其中高(H組)中(M組)劑量組創(chuàng)面愈合率明顯低于低(L組)劑量組(P0.05)。 巨噬細(xì)胞數(shù)量:高(H)、中(M)、低(L)劑量組第7天創(chuàng)面肉芽組織巨噬細(xì)胞數(shù)量明顯少于對照組(C)。 結(jié)論香煙煙霧導(dǎo)致創(chuàng)面愈合延遲,固定時間創(chuàng)面愈合率降低,創(chuàng)面肉芽組織巨噬細(xì)胞數(shù)量減少。
[Abstract]:PurposeTo observe the effect of cigarette smoke on wound healing time of rats and the effect of fixed time wound healing rate in each group the number of macrophages in wound healing process was counted by immunohistochemical method with CD68 monoclonal antibody.To explore the possible effects of macrophages on wound healing.MethodForty healthy adult SD rats, weighing 190-260 g, were randomly divided into 4 groups: control group (C), low tar content group (L group), middle tar content group (M group) and high tar content group (H group) with 10 rats in each group.In each group, 4 circles with 2cm diameter were drawn after hair removal on the back skin of each group. The whole skin layer was cut out according to the circle drawn, and 4 equal size circular wounds were formed.The distance of each wound was larger than that of 2cm.C group without any intervention. The rats in group H were placed in a self-made fumigation box (30L). The rats were fumigated with 0. 5 mg 0. 5 mg of 0. 5 mg of 0. 8 mg of tar and smoked for 30 minutes every day until the wound healed.The wound healing time was observed, and the wound healing rate was observed on the 7th and 16th days, respectively. The tissue around the wound was taken on the 3rd day and the 7th day, and the number of macrophages was counted by immunohistochemical method and CD68 labeled by monoclonal antibody.ResultWound healing time: the healing time of the high dose group and the low dose group was significantly longer than that of the control group (P 0.05).It was suggested that the high, medium and low concentration of tar could prolong the healing time of wound.The healing time of high and middle tar concentration group was longer than that of low tar content group.Wound healing rate: compared with the control group (C group), the wound healing rate of the group with high Tar content, group M, and group L) was lower than that of the control group on day 716, and the rate of wound healing was lower than that of the control group on day 716.The wound healing rate in group M was significantly lower than that in group L (P 0.05).Number of macrophages: the number of macrophages in the granulation tissue of the high dose group was significantly lower than that of the control group on the 7th day.Conclusion cigarette smoke can delay wound healing, decrease wound healing rate and decrease the number of granulation macrophages.
