【摘要】：背景:研究發(fā)現(xiàn)粒細胞集落刺激因子可以用于治療粒細胞減少癥,關(guān)于粒細胞集落刺激因子治療膿毒癥的研究不多。目的:探討重組人粒細胞巨噬細胞集落刺激因子對膿毒癥小鼠的治療作用。方法:采用腹腔內(nèi)注射脂多糖的方法建立膿毒癥小鼠動物模型,建模后6,30 h皮下注射重組人粒細胞巨噬細胞集落刺激因子,觀察小鼠的肺組織病理變化、外周血中性粒細胞CD64表達以及血清腫瘤壞死因子α、白細胞介素10水平。結(jié)果與結(jié)論:(1)治療后小鼠支氣管上皮尚完整,間質(zhì)輕度增生,見少量中性粒細胞浸潤;(2)治療后1,3,7 d治療組小鼠外周血中性粒細胞CD64的表達和血清白細胞介素10水平顯著高于模型組和正常對照組(P0.05);(3)治療后1,3,7 d治療組小鼠血清腫瘤壞死因子α水平明顯高于正常對照組(P0.05),但低于模型組(P0.05);(4)結(jié)果表明,重組人粒細胞巨噬細胞集落刺激因子具有增強膿毒癥小鼠中性粒細胞功能和抗炎作用。
[Abstract]:Background: it has been found that granulocyte colony stimulating factor can be used to treat granulocytopenia, but few studies have been done on the treatment of sepsis with granulocyte colony stimulating factor.Aim: to investigate the therapeutic effect of recombinant human granulocyte macrophage colony stimulating factor (RHGF) on sepsis mice.Methods: an animal model of sepsis was established by intraperitoneal injection of lipopolysaccharide. Recombinant human granulocyte macrophage colony stimulating factor was injected subcutaneously at 6h after modeling and the pathological changes of lung tissue were observed.The expression of CD64 in peripheral blood neutrophils and the levels of tumor necrosis factor 偽 and interleukin 10 in serum.Results and conclusion: after treatment, the bronchial epithelium of mice was intact and the interstitial hyperplasia was mild.The expression of CD64 in peripheral blood neutrophils and the level of serum interleukin 10 in the treatment group were significantly higher than those in the model group and the normal control group on the 7th day after treatment.The level of tumor necrosis factor 偽 was significantly higher than that of the normal control group (P 0.05), but lower than that of the model group (P 0.05).Recombinant human granulocyte macrophage colony stimulating factor can enhance neutrophil function and anti-inflammatory effect in septic mice.
