【摘要】：目的探討右美托咪定對膿毒血癥大鼠腎功能及緊密連接蛋白表達的影響。方法 SD大鼠30只,隨機分為右美托咪定處理組(Dex組)、膿毒血癥模型組(M組)、生理鹽水對照組(C組),每組10只。Dex組大鼠靜脈注射脂多糖(LPS)5 mg/kg,之后泵注Dex 7μg/kg,15 min后調(diào)整劑量為5μg/kg·h,持續(xù)泵注30 min;模型組按同樣方法給予LPS后泵注生理鹽水;C組始終采用上述方法給予生理鹽水。24 h后取血液分離血漿,多功能生化儀檢測肌酐(Scr)和尿素氮(BUN)含量;ELISA檢測IL-1β和TNF-α含量;Western blot檢測腎組織中緊密連接蛋白ZO-1和occludin的表達變化。結(jié)果與C組相比,模型組損傷較重,可見大量炎性細胞浸潤,有腎小球腎炎表現(xiàn),而Dex組未見明顯的炎癥細胞,細胞異常較少,與模型組相比,損傷減輕。腎功能及炎性因子檢測結(jié)果顯示,與C組比較,各組血漿中Scr、BUN、IL-1β和TNF-α含量均顯著升高(P0.05),ZO-1和occludin蛋白表達水平均下降(P0.05);與模型組比較,Dex組各項指標均明顯降低(P0.05),ZO-1和occludin蛋白表達水平均升高(P0.05)。結(jié)論右美托咪定可改善膿毒血癥所致的急性腎損傷的腎功能,抑制炎癥反應(yīng),提升緊密連接蛋白,對腎臟有一定的保護作用。
[Abstract]:Objective to investigate the effect of dexmedetomidine on renal function in rats with sepsis and the expression of tight junction protein. Methods 30 SD rats were randomly divided into dexmedetomidine group (Dex group), sepsis model group (M group), saline control group (C group), 10 rats in each group.Dex group of rats by intravenous injection of lipopolysaccharide (LPS) after infusion of Dex 5 mg/kg, 7 g/kg, 15 min after adjusting the dose of 5 g/kg / h, continuous infusion of 30 min; model group were given LPS after infusion of normal saline in the same way; C group always adopts the method given saline.24 after h the blood plasma separation, multifunctional biochemical detection of serum creatinine (Scr) and urea nitrogen (BUN) content; ELISA detection of IL-1 beta and TNF- alpha content; the expression of tight junction protein ZO-1 and occludin detection of Western blot in renal tissue. Results compared with C group, the model group had more severe injury, there are a lot of inflammatory cells infiltration of glomerular nephritis table Now, while the Dex group had no obvious inflammatory cells, abnormal cells less, compared with the model group. Reduce the damage of renal function and inflammatory factor test results showed that compared with C group, plasma Scr, BUN, IL-1 beta and TNF- alpha were significantly increased (P0.05), the expression level of ZO-1 and occludin protein decreased (P0.05); compared with the model group, the indexes of group Dex were significantly lower (P0.05), the expression level of ZO-1 and occludin protein were increased (P0.05). Conclusion dexmedetomidine can improve sepsis induced acute kidney injury renal function, inhibit inflammatory reaction, enhance the tight junction protein, protection effects on the kidney.
