日本血吸蟲表皮生長因子受體基因的鑒定和功能研究(英文)

目的鑒定日本血吸蟲表皮生長因子受體基因(SjEGFR),并闡明其在日本血吸蟲生長和生殖發(fā)育中的作用。方法通過5’-RACE和3’-RACE擴(kuò)增、測序獲取SjEGFR基因全長片段。采用熒光定量PCR(qPCR)技術(shù)檢測該基因在日本血吸蟲不同發(fā)育階段(16、18、20、22、24、26 d)表達(dá)情況。使用整條蟲體原位雜交法對感染24 d的日本血吸蟲雌、雄蟲SjEGFR基因轉(zhuǎn)錄本進(jìn)行定位。通過體內(nèi)持續(xù)RNA干擾(RNAi)技術(shù)特異性敲低SjEGFR基因,待蟲體發(fā)育至30 d收集蟲體。統(tǒng)計(jì)分析雌、雄蟲回收數(shù)量以及蟲體長度,通過明礬卡紅染色觀察蟲體生長發(fā)育情況。結(jié)果首次鑒定了SjEGFR基因,其結(jié)構(gòu)具有一個保守的酪氨酸激酶位點(diǎn)。整條蟲體原位雜交定位顯示該基因轉(zhuǎn)錄本在蟲體各處均有表達(dá),在雌、雄蟲腸道部位呈現(xiàn)出明顯著色的高表達(dá)。體內(nèi)RNAi顯示,SjEGFR基因表達(dá)被特異性敲低后,雌、雄蟲生長受阻。RNAi組雌蟲子宮內(nèi)無蟲卵,腸道內(nèi)無色素沉積、卵黃腺發(fā)育遲滯、卵巢發(fā)育受阻;雄蟲呈現(xiàn)睪丸個數(shù)減少、體積變小,出現(xiàn)了更多未成熟的精母細(xì)胞。結(jié)論特異性敲低SjEGFR基因表達(dá)可影響日本血吸蟲生長和生殖發(fā)育,阻...
