人牙齦間充質(zhì)干細(xì)胞對(duì)B細(xì)胞的作用及機(jī)制研究

目的探討人牙齦間充質(zhì)干細(xì)胞(GMSC)對(duì)B細(xì)胞增殖與分化的調(diào)節(jié)功能及其相關(guān)分子機(jī)制。方法分離提取GMSC,從外周血分選出B細(xì)胞。將GMSC或纖維母細(xì)胞分別與B細(xì)胞進(jìn)行體外共培養(yǎng),設(shè)為GMSC組和纖維母細(xì)胞組,檢測兩組B細(xì)胞的擴(kuò)增情況;檢測兩組細(xì)胞上清中IgG1與IgM的表達(dá)情況;檢測兩組細(xì)胞白細(xì)胞介素(IL)-6、Perforin、干擾素(IFN)-γ和腫瘤壞死因子(TNF)-α的分泌情況;檢測兩組B細(xì)胞表達(dá)IL-10與轉(zhuǎn)化生長因子(TGF)-β的水平;檢測兩組B細(xì)胞中PC-1的表達(dá)情況;檢測GMSC調(diào)控B細(xì)胞功能的信號(hào)通路;檢測GMSC對(duì)B細(xì)胞活化T細(xì)胞功能的調(diào)控。結(jié)果與纖維母細(xì)胞組比較,GMSC組B細(xì)胞的擴(kuò)增明顯減弱(P<0.05),GMSC與B細(xì)胞共培養(yǎng)后,能明顯抑制B細(xì)胞分泌IgG1和IgM,明顯抑制IL-6、Perforin、IFN-γ和TNF-α的分泌(均為P<0.05)。與纖維母細(xì)胞組比較,GMSC組中的IL-10與TGF-β分泌量更高(均為P<0.05)。GMSC組中PC-1的表達(dá)水平明顯降低(P<0.05)。加入TGF-β受體抑制劑ALK5后,...
