NGF在大鼠慢性偏頭痛模型中的表達及機制研究

背景:我們前期研究發(fā)現(xiàn),三叉神經(jīng)脊束尾核(trigeminal nucleus caudalis,TNC)中酸敏感離子通道3(acid-sensing ion channel 3,ASIC3)參與了反復發(fā)作性偏頭痛的病理過程。神經(jīng)生長因子(nerve growth factor,NGF)可以誘導許多疼痛疾病的痛覺敏化,并且可以調(diào)控ASIC3的表達。然而,NGF在慢性偏頭痛(chronic migraine,CM)中的作用尚不清楚。所以我們探究了CM大鼠TNC部位NGF的表達,NGF對ASIC3的調(diào)控作用,及相關(guān)的信號通路在炎性湯(inflammatory soup,IS)誘導的皮膚痛覺過敏中的作用。方法:1.通過硬腦膜反復滴注7日IS建立CM大鼠模型,模擬CM的發(fā)作。von Frey測定大鼠眶周及足底的痛閾值,以此評價模型的建立。2.利用PCR,Western blot檢測TNC部位NGF m RNA水平及蛋白水平,免疫熒光檢測NGF在TNC神經(jīng)元中的定位情況。3.CM大鼠側(cè)腦室注射NGF中和抗體(anti-NGF neutralizing antibody,anti-NGF),von...
