Lrrc34在精原干細胞中高表達,且為體外精原干細胞增殖的必需因子(英文)

目的探討維持精原干細胞及其干細胞特性的關(guān)鍵基因及亮氨酸重復(fù)序列(LRR)家族成員Lrrc34在小鼠精原干細胞中的功能。方法應(yīng)用差異表達基因(DEG)分析、基因本體(GO)分析和京都基因和基因組百科全書(KEGG)分析等生物信息學(xué)方法尋找潛在的多能性相關(guān)基因。分別采用逆轉(zhuǎn)錄聚合酶鏈式反應(yīng)(RT-PCR)、免疫熒光(IF)檢測Lrrc34在小鼠精原干細胞中的m RNA和蛋白質(zhì)表達水平。采用RNA干擾實驗研究Lrrc34敲低后對體外培養(yǎng)精原干細胞的瞬時效應(yīng)。結(jié)果我們分別在ID4-EGFP+(G)和ID4-EGFP+/TSPAN8High (TH)、G和ID4-EGFP+/TSPAN8Low (TL)、TH和TL組發(fā)現(xiàn)了8、8、11個差異表達基因,并在這些差異表達基因組成的相互作用網(wǎng)絡(luò)中發(fā)現(xiàn)了11個顯著的相互作用模塊。第五個相互作用模塊中與纖維母細胞生長因子2存在相互作用的差異表達基因Lrrc34被選擇作為多能性相關(guān)候選基因。免疫熒光實驗顯示Lrrc34特異表達于被LIN28A標(biāo)記的未分化精原細胞的部分亞群中,RT-PCR證明了Lrrc34高表達于出生后7天和成年小鼠的精原干細胞中。Lrrc3...
