四川呼吸道合胞病毒F蛋白基因序列和抗原表位變異分析

【摘要】:目的分析呼吸道合胞病毒(RSV)四川分離株F蛋白基因序列,探討F蛋白基因和抗原表位變異情況。方法 RT-PCR方法擴(kuò)增F蛋白近全長(zhǎng)序列并測(cè)序,分析核苷酸和氨基酸變異位點(diǎn),比較不同亞型和基因型間中和表位、細(xì)胞毒性T淋巴細(xì)胞(CTL)表位基因組成。結(jié)果 10株RSV F蛋白的核苷酸和氨基酸P-distance分別為0.102±0.005和0.058±0.006,中和表位47F和L4在亞型間高度保守,而RS-348和7C2在亞型內(nèi)保守,CTL表位HLA B*57、HLA A*01和HLA Cw*12也是亞型內(nèi)高度保守,亞型間存在個(gè)別氨基酸變異。結(jié)論 RSV四川分離株F蛋白基因保守度較高,抗原表位在亞型內(nèi)保守,已在A亞型上識(shí)別的CTL表位可能在B亞型內(nèi)不能識(shí)別。
[Abstract]:Objective to analyze the sequence of F protein gene of respiratory syncytial virus (RSV) Sichuan isolate and to explore the variation of F protein gene and antigenic epitope. Methods the near-full-length sequence of F protein was amplified and sequenced by RT-PCR. The variation sites of nucleotides and amino acids were analyzed. The neutralization epitopes among different subtypes and genotypes and the (CTL) epitope gene composition of cytotoxicity T lymphocytes were compared.
