RANKL和M-CSF在CIA大鼠關(guān)節(jié)滑膜的表達(dá)及藥物的干預(yù)作用

【摘要】:目的初步探討RANKL和M-CSF在CIA大鼠關(guān)節(jié)滑膜的表達(dá)及T-614、MTX的干預(yù)作用。方法通過(guò)雞Ⅱ型膠原和完全弗氏佐劑混合的膠原乳劑免疫大鼠,建立CIA大鼠模型,將CIA大鼠隨機(jī)分為模型對(duì)照組、T-614組、MTX組和T-614+MTX組。藥物干預(yù)組大鼠分別用T-614、MTX及T-614聯(lián)合MTX干預(yù)3周。關(guān)節(jié)炎指數(shù)(arthritis index,AI)評(píng)分評(píng)價(jià)關(guān)節(jié)炎程度,HE染色觀察踝關(guān)節(jié)組織形態(tài)學(xué)變化,X線片觀察骨侵蝕情況,高頻超聲觀察踝關(guān)節(jié)并測(cè)量滑膜厚度,Western blot法檢測(cè)滑膜RANKL和M-CSF表達(dá)。結(jié)果與正常對(duì)照組相比,模型對(duì)照組AI評(píng)分、病理評(píng)分、X線評(píng)分、超聲下滑膜厚度、關(guān)節(jié)滑膜RANKL,M-CSF表達(dá)均增加(P0.05);與模型對(duì)照組相比,T-614組、MTX組、T-614聯(lián)合MTX組上述觀察指標(biāo)均降低(P0.05);與T-614組、MTX組相比,T-614聯(lián)合MTX組上述觀察指標(biāo)均降低(P0.05);T-614組和MTX組之間上述觀察指標(biāo)差異無(wú)統(tǒng)計(jì)學(xué)意義(P0.05)。結(jié)論 RANKL和M-CSF參與了CIA大鼠關(guān)節(jié)滑膜病變的發(fā)生、發(fā)展,T-614、MTX可能通過(guò)減少細(xì)胞因子RANKL,M-CSF的生成來(lái)延緩關(guān)節(jié)滑膜病變的進(jìn)展,T-614與MTX具有一定的協(xié)同效應(yīng)。
[Abstract]:Objective To study the expression of RANKL and M-CSF in the joint synovium of CIA rats and the intervention of T-614 and MTX. Methods The CIA rats were randomly divided into the model control group, the T-614 group, the MTX group and the T-614 + MTX group by using the collagen emulsion mixed with the chicken type II collagen and the complete Freund's adjuvant to immunize the rat. The rats of the drug intervention group were treated with T-614, MTX and T-614 in combination with MTX for 3 weeks. Arthritis index (AI) score was used to evaluate the degree of arthritis, and the morphological changes of the ankle tissue were observed with HE staining. The bone erosion was observed by X-ray. The thickness of the synovium was measured by high-frequency ultrasound, and the expression of RANKL and M-CSF was detected by Western blot. Results Compared with the normal control group, the AI score, the pathological score, the X-ray score, the ultrasonic glide film thickness, the joint synovial RANKL and the M-CSF expression in the model control group were all increased (P0.05), and the above-mentioned observation indexes of the T-614 group, the MTX group and the T-614 combined MTX group were all decreased (P0.05). Compared with the T-614 group and the MTX group, the above-mentioned observation index of the T-614 combined MTX group was decreased (P0.05), and the difference of the observation indexes between the T-614 group and the MTX group was not statistically significant (P0.05). Conclusion RANKL and M-CSF are involved in the development, development, T-614 and MTX of the joint synovium of CIA rats.
