【摘要】：利用軟體組織破碎過(guò)濾法提取湖北釘螺(Oncomelania hupensis)血淋巴細(xì)胞,加入檸檬酸葡萄糖溶液作為抗凝劑,經(jīng)吉氏染色和美蘭染色后,用Motic B5計(jì)算機(jī)成像系統(tǒng)觀察其形態(tài)。提取單只釘螺的血淋巴細(xì)胞,統(tǒng)計(jì)細(xì)胞數(shù)。結(jié)果共觀察54個(gè)血淋巴細(xì)胞,可見(jiàn)3種形態(tài):圓形有顆粒大細(xì)胞,直徑(21.59～31.97)μm,細(xì)胞核呈卵圓形,有顆粒,核質(zhì)比0.38±0.08,約占細(xì)胞總數(shù)35.95%(19/54);圓形無(wú)顆粒小細(xì)胞,直徑(13.24～20.77)μm,細(xì)胞核呈卵圓形,無(wú)顆粒,核質(zhì)比0.38±0.90,約占細(xì)胞總數(shù)51.63%(28/54);梭形細(xì)胞,大小(17.60～25.47)μm×(27.19～30.25)μm,細(xì)胞核呈長(zhǎng)橢圓形,無(wú)顆粒,核質(zhì)比0.44±0.20,約占細(xì)胞總數(shù)12.42%(7/54)。用過(guò)濾法提取單只釘螺血淋巴細(xì)胞均數(shù)約為19.46萬(wàn)個(gè)。該法是提取釘螺血淋巴細(xì)胞的一種有效方法。
[Abstract]:The blood lymphocytes of (Oncomelania hupensis) of Oncomelania hupensis were extracted by soft tissue fragmentation and filtration. The blood lymphocytes of Oncomelania hupensis were treated with citric acid and glucose solution as anticoagulant. After Ji's staining and Meilan staining, the morphology of the lymphocytes was observed by Motic B5 computer imaging system. The blood lymphocytes of a single Oncomelania hupensis were extracted and the number of cells was counted. Results A total of 54 blood lymphocytes were observed.
