NIRF可抑制乙型肝炎病毒在水動力法轉(zhuǎn)染HBV小鼠模型中的復(fù)制及表達(dá)

【摘要】:目前對NIRF(Np95/ICBP-90 like RING finger protein)的研究正朝著細(xì)胞癌變進(jìn)程以及表觀遺傳學(xué)的方向發(fā)展,但在體內(nèi)NIRF對乙型肝炎病毒(HBV)的復(fù)制及表達(dá)的影響,目前尚不明確.通過高壓水動力法轉(zhuǎn)染HBV小鼠模型,不同時間點收集血液和肝組織標(biāo)本,熒光定量PCR檢測血清及肝組織中病毒載量,WesternBlot檢測肝組織HBc(hepatitis B virus core protein)表達(dá),ELISA檢測血清HBeAg表達(dá),并通過免疫組化染色檢測HBsAg、HBcAg在肝組織中的定位及表達(dá).小鼠轉(zhuǎn)染pAAV-HBV1.3和NIRF以后,血清及肝組織病毒載量降低(n=3,P0.05),HBc蛋白及HBV相關(guān)抗原的表達(dá)受到抑制,說明在水動力法轉(zhuǎn)染HBV小鼠模型中NIRF對HBV的復(fù)制及表達(dá)起到抑制作用,期待能為后續(xù)的HBV致病機(jī)理及治療藥物的研究與開發(fā)提供支持與幫助.

The viral load in serum and liver tissue was detected by fluorescence quantitative PCR, the expression of HBc (hepatitis B virus core protein) in liver tissue was detected by WesternBlot, and the expression of HBeAg in serum was detected by ELISA.
