Tsp10多肽免疫血清在體外對(duì)旋毛蟲侵入腸上皮細(xì)胞及幼蟲發(fā)育的影響

【摘要】:目的觀察Tsp10多肽免疫血清在體外對(duì)旋毛蟲侵入腸上皮細(xì)胞及幼蟲發(fā)育的影響。方法將不同免疫血清與含有旋毛蟲腸道感染性幼蟲的半固體培養(yǎng)基混合后,接種至正常小鼠腸上皮細(xì)胞(intestinal epithelial cells,IEC)單層上,37℃5%CO2培養(yǎng)2h、36h時(shí)分別觀察幼蟲侵入率、細(xì)胞單層受損面積及發(fā)育至Ⅱ~Ⅳ期幼蟲的百分比;免疫熒光試驗(yàn)檢測(cè)受損腸上皮細(xì)胞中的旋毛蟲抗原。結(jié)果幼蟲在半固體培養(yǎng)基培養(yǎng)2h時(shí),Tsp10多肽免疫血清組的幼蟲侵入率與細(xì)胞單層受損面積(46.3%,24.3mm2)均明顯低于T7噬菌體免疫血清組(94.1%,65.3mm2)及正常小鼠血清組(92.5%,65.7mm2)(P0.01);培養(yǎng)36h時(shí),Tsp10多肽免疫血清組Ⅱ~Ⅳ期幼蟲的百分比(16%)亦明顯低于T7噬菌體免疫血清組(37%)及正常小鼠血清組(38%)(P0.01)。Tsp10多肽及ES抗原免疫血清的免疫熒光試驗(yàn)結(jié)果顯示,受損的IEC呈亮綠色熒光,而應(yīng)用T7噬菌體免疫血清及正常小鼠血清時(shí)則為陰性。結(jié)論 Tsp10多肽免疫血清可阻止部分旋毛蟲幼蟲對(duì)腸上皮細(xì)胞的侵入與發(fā)育,Tsp10可能是旋毛蟲的主要侵入蛋白之一。
