非同源腦脊液誘導(dǎo)非同源間充質(zhì)干細(xì)胞分化為神經(jīng)干細(xì)胞的對(duì)比研究

【摘要】:尋找一種符合患者治療需求的干細(xì)胞獲取模式,從而進(jìn)一步推動(dòng)神經(jīng)干細(xì)胞廣泛應(yīng)用于臨床。本實(shí)驗(yàn)采用志愿者、患者腦脊液體外誘導(dǎo)異體臍血源間充質(zhì)干細(xì)胞(UMSCs)和患者骨髓源間充質(zhì)干細(xì)胞(BMSCs)分化為神經(jīng)干細(xì)胞,并采用形態(tài)學(xué)、流式細(xì)胞分析、免疫組化、熒光免疫組化法等方法對(duì)誘導(dǎo)后的細(xì)胞進(jìn)行簽定;同時(shí),記錄各誘導(dǎo)組獲得臨床可用神經(jīng)干細(xì)胞所需的時(shí)間及細(xì)胞數(shù)計(jì)數(shù)。結(jié)果證實(shí)各組均可使間充質(zhì)干細(xì)胞(MSCs)定向分化為神經(jīng)干細(xì)胞,所獲得的細(xì)胞數(shù)亦可滿足臨床需要,但在所需的天數(shù)及具體獲得細(xì)胞數(shù)量上各組間有一定差異,其中以患者BMSCs用自愿者腦脊液誘導(dǎo)組所獲得神經(jīng)干細(xì)胞計(jì)數(shù)最高,且其所需時(shí)間最短。
[Abstract]:Search for a stem cell acquisition model that meets the needs of patients, and further promote the wide application of neural stem cells in clinical practice. In this experiment, (UMSCs) from cord blood derived mesenchymal stem cells and (BMSCs) from bone marrow derived mesenchymal stem cells were induced to differentiate into neural stem cells from volunteers and cerebrospinal fluid (CSF) in vitro. Morphology, flow cytometry and immunohistochemistry were used. The induced cells were identified by fluorescence immunohistochemical method. At the same time, the time and count of the number of neural stem cells were recorded in each induction group. The results showed that the (MSCs) of mesenchymal stem cells could be differentiated into neural stem cells in each group, and the number of cells obtained could also meet the clinical needs, but there were some differences in the number of specific cells and the number of cells obtained in each group. The number of neural stem cells was the highest in the group of patients with BMSCs induced by cerebrospinal fluid (CSF), and it took the shortest time.
