剛地弓形蟲親環(huán)蛋白基因編碼蛋白主要特性與抗原表位的生物信息學(xué)分析

【摘要】:目的應(yīng)用生物信息學(xué)方法分析剛地弓形蟲親環(huán)蛋白(cyclophilin protein,TgCyP)基因編碼蛋白的主要特性與抗原表位,篩選該蛋白的T/B細胞聯(lián)合表位。方法利用蛋白分析專家系統(tǒng)(ExPASy)提供的ProtParam、SignalP、SOSUI、SOPMA、PSORT、MotifScan和NCBI上的ORF finder、Bcepred、SYFPEITHI等生物信息學(xué)在線分析程序,結(jié)合Gene Runner、DNAMAN等生物信息學(xué)軟件,分析和預(yù)測TgCyP蛋白的理化性質(zhì)、信號肽、可溶性、表面可及性、可塑性,跨膜區(qū),翻譯后修飾位點、親(疏)水性、二級結(jié)構(gòu)、亞細胞定位、T/B細胞抗原表位等。結(jié)果該蛋白由179個氨基酸組成,分子式為C884H1378N242O258S4,分子質(zhì)量單位為19.6523ku,等電點理論值為6.06,波長280nm時的吸光度(A)值為0.787,半衰期為30h,有4個表面可及性參數(shù)≥1.9的區(qū)域、4個親水性參數(shù)得分≥1.9的區(qū)域、7個柔韌性參數(shù)得分≥2的區(qū)域、9個翻譯后修飾位點、8個潛在B細胞表位、3個T/B細胞聯(lián)合表位,為可溶性表達蛋白。結(jié)論 TgCyP蛋白為可溶性表達蛋白,有3個T/B細胞聯(lián)合表位,具有潛在的免疫保護作用。
[Abstract]:Objective to analyze the main characteristics and antigenic epitopes of cyclophile protein (cyclophilin protein,TgCyP) gene of Toxoplasma gondii by bioinformatics, and to screen the T / B cell epitopes of Toxoplasma gondii (Toxoplasma gondii).
