【摘要】：目的應(yīng)用生物信息學(xué)方法分析剛地弓形蟲(chóng)親環(huán)蛋白(cyclophilin protein,TgCyP)基因編碼蛋白的主要特性與抗原表位,篩選該蛋白的T/B細(xì)胞聯(lián)合表位。方法利用蛋白分析專(zhuān)家系統(tǒng)(ExPASy)提供的ProtParam、SignalP、SOSUI、SOPMA、PSORT、MotifScan和NCBI上的ORF finder、Bcepred、SYFPEITHI等生物信息學(xué)在線(xiàn)分析程序,結(jié)合Gene Runner、DNAMAN等生物信息學(xué)軟件,分析和預(yù)測(cè)TgCyP蛋白的理化性質(zhì)、信號(hào)肽、可溶性、表面可及性、可塑性,跨膜區(qū),翻譯后修飾位點(diǎn)、親(疏)水性、二級(jí)結(jié)構(gòu)、亞細(xì)胞定位、T/B細(xì)胞抗原表位等。結(jié)果該蛋白由179個(gè)氨基酸組成,分子式為C884H1378N242O258S4,分子質(zhì)量單位為19.6523ku,等電點(diǎn)理論值為6.06,波長(zhǎng)280nm時(shí)的吸光度(A)值為0.787,半衰期為30h,有4個(gè)表面可及性參數(shù)≥1.9的區(qū)域、4個(gè)親水性參數(shù)得分≥1.9的區(qū)域、7個(gè)柔韌性參數(shù)得分≥2的區(qū)域、9個(gè)翻譯后修飾位點(diǎn)、8個(gè)潛在B細(xì)胞表位、3個(gè)T/B細(xì)胞聯(lián)合表位,為可溶性表達(dá)蛋白。結(jié)論 TgCyP蛋白為可溶性表達(dá)蛋白,有3個(gè)T/B細(xì)胞聯(lián)合表位,具有潛在的免疫保護(hù)作用。
[Abstract]:Objective to analyze the main characteristics and antigenic epitopes of cyclophile protein (cyclophilin protein,TgCyP) gene of Toxoplasma gondii by bioinformatics, and to screen the T / B cell epitopes of Toxoplasma gondii (Toxoplasma gondii). Methods Bioinformatics analysis programs such as ProtParam,SignalP,SOSUI,SOPMA,PSORT,MotifScan and ORF finder,Bcepred,SYFPEITHI on NCBI provided by protein analysis expert system (ExPASy) and bioinformatics software such as Gene Runner,DNAMAN were used. Results the protein was composed of 179 amino acids, the molecular formula was C884H1378N242O258S4, the unit of molecular mass was 19.6523ku. the theoretical value of isoelectric point was 6.06. the absorbance (A) value of wavelength 280nm was 0.787, and the half-life was 30 hours.
