低氧對大鼠大腦皮層神經(jīng)干細胞增殖和分化的影響

【摘要】:目的:觀察和分析不同低氧濃度對大鼠大腦皮層神經(jīng)干細胞(neural stem cells,NSC S)增殖和分化的影響。方法:在1%O2、4%O2和常氧環(huán)境(20%O2)下培養(yǎng)新生SD大鼠神經(jīng)干細胞,對神經(jīng)球計數(shù)并測量其直徑,分析不同低氧對神經(jīng)干細胞增殖效率和增殖速度的影響;分化48 h后行β微管蛋白Ⅲ(β-tubulin III)和膠質纖維酸性蛋白(glial fibrillary acidic protein,GFAP)免疫細胞熒光染色,對神經(jīng)元和星形膠質細胞進行計數(shù)并分別計算各占的比例。結果:(1)低氧對神經(jīng)干細胞增殖效率和增殖速度的影響:1%低氧組神經(jīng)球數(shù)量明顯低于常氧對照組,具有顯著性差異(P0.05);4%低氧組神經(jīng)球數(shù)量高于對照組,具有顯著性差異(P0.05)。在24 h、36 h和48 h,1%低氧組神經(jīng)球的直徑都小于對照組,在24 h和36 h時具有顯著性差異(P0.05),在48 h具有非常顯著性差異(P0.01);在24 h、36 h和48 h,4%低氧組神經(jīng)球的直徑都大于對照組,在36 h時具有顯著性差異(P0.05),在48 h具有非常顯著性差異(P0.01)。(2)1%低氧組神經(jīng)元的比例(0.614±0.056)顯著高于其對照組(0.471±0.067)(P0.01);而1%低氧組星形膠質細胞的比例(0.241±0.051)低于其對照組(0.322±0.067)(P0.05)。4%低氧組神經(jīng)元的比例(0.625±0.072)顯著高于其對照組(0.513±0.032)(P0.05);而4%低氧組星形膠質細胞的比例(0.237±0.053)顯著低于其對照組(0.285±0.042)(P0.05)。結論:和常氧對照組相比,4%O2促進大鼠大腦皮層神經(jīng)干細胞的增殖,而1%O2抑制了神經(jīng)干細胞的增殖;1%O2和4%O2都促進神經(jīng)干細胞向神經(jīng)元方向分化,同時抑制向星形膠質細胞分化,1%O2的這種作用更為顯著。
