骨形態(tài)發(fā)生蛋白與血小板衍生性生長因子在骨形成中協(xié)同作用的研究進展

【摘要】:骨形成是成骨細胞和破骨細胞在多種因子存在的微環(huán)境中共同作用的結果。骨損傷早期,控制骨祖細胞誘導作用的各種生物化學因素和生物生理學因素起著重要作用,然而在后期促進骨改建的各種細胞因子起主要作用。新近研究表明:各生長因子通過刺激成骨細胞的增殖及其活性,調節(jié)局部骨形成作用,然而關于生長因子在調節(jié)成骨細胞代謝中的相互作用關系尚不完全清楚。在眾多骨生長因子中,骨形態(tài)發(fā)生蛋白(BMPs)被公認是目前最強的骨誘導因子,而血小板衍生性生長因子(PDGF)對骨改建有雙向調節(jié)作用,因此,進一步探究BMP和PDGF及其二者在骨形成中的作用意義深遠。本文作者探索BMPs和PDGF的化學結構及生物學作用,闡述BMP與PDGF在骨形成中協(xié)調作用的明確性和復雜性。
[Abstract]:Bone formation is the result of the interaction of osteoblasts and osteoclasts in the microenvironment in which many factors exist. In the early stage of bone injury, various biochemical and biophysiological factors controlling the induction of bone progenitor cells play an important role. However, various cytokines that promote bone remodeling play a major role in the later stage of bone injury. Recent studies have shown that the growth factors regulate local bone formation by stimulating the proliferation and activity of osteoblasts, but the interaction of growth factors in the regulation of osteoblast metabolism is not completely clear. Among the many bone growth factors, bone morphogenetic protein (BMPs) is recognized as the strongest bone inducer at present, while platelet-derived growth factor (PDGF) has a bidirectional effect on bone remodeling. To further explore the role of BMP and PDGF in bone formation is of great significance. The authors explore the chemical structure and biological functions of BMPs and PDGF, and explain the clarity and complexity of the coordinated role of BMP and PDGF in bone formation.
