生物鐘基因Rev-erb-α缺失導(dǎo)致雌鼠生理周期延長(zhǎng)

【摘要】:在哺乳動(dòng)物中,生物節(jié)律與生殖生理和生殖內(nèi)分泌密切相關(guān),流行病學(xué)研究顯示長(zhǎng)期倒時(shí)差的女性表現(xiàn)出生殖力低下和不良妊娠的概率更高,但是人們對(duì)生物節(jié)律影響生殖的機(jī)制了解還很少。本研究旨在以時(shí)鐘基因Rev-erb-α敲除型(Rev-erb-α-/-)雌性小鼠為模型初步探索生物節(jié)律對(duì)生殖的影響。將Rev-erb-α-/-和野生型(Rev-erb-α+/+)小鼠與C57BL/6J小鼠交配觀察生殖力變化,用蘇木精-伊紅(HE)染色法觀察卵巢組織變化,將Rev-erb-α+/+與Rev-erb-α-/-雌性小鼠分別與C57BL/6J雄性小鼠交配后比較見(jiàn)栓后1.5d統(tǒng)計(jì)的胚胎數(shù)量,用陰道涂片結(jié)合周期圖和周期譜觀察小鼠生理周期變化。結(jié)果顯示,Rev-erb-α-/-雌性小鼠與C57BL/6J雄鼠交配是不生育的,并且Rev-erb-α-/-雌性小鼠生成的胚胎數(shù)量明顯減少。Rev-erb-α-/-雌性小鼠卵巢存在各級(jí)卵泡,并且結(jié)構(gòu)與野生型(Rev-erb-α+/+)雌性小鼠相比無(wú)明顯差異。Rev-erb-α-/-雌性小鼠的生理周期較Rev-erb-α+/+小鼠周期長(zhǎng)約3.22d,較Rev-erb-α+/+雌性小鼠延長(zhǎng)63.5%。以上結(jié)果提示,生物鐘基因Rev-erb-α缺失導(dǎo)致雌性小鼠生理周期明顯延長(zhǎng),可能是影響雌性小鼠生育的原因之一。因此,生物節(jié)律的穩(wěn)定對(duì)于生理周期具有重要作用。
[Abstract]:In mammals, biological rhythms are closely related to reproductive physiology and reproductive endocrine. Epidemiological studies have shown that women with chronic jet lag are more likely to have low fertility and poor pregnancy. But little is known about the mechanisms that biological rhythms affect reproduction. The aim of this study was to explore the effects of biological rhythm on reproduction in Rev-erb- 偽 -r- female mice with clock gene Rev-erb- 偽 knockout (Rev-erb- 偽 -r-). Rev-erb- 偽 -r- and wild type (Rev-erb- 偽 /) mice were mated with C57BL/6J mice to observe the change of fecundity, and the changes of ovarian tissue were observed by hematoxylin-eosin (HE) staining. After mating with C57BL/6J male mice, Rev-erb- 偽 / Rev-erb- 偽 -r-female mice were compared with the number of embryos counted 1.5 days after embolization. The physiological cycle changes of mice were observed by vaginal smear combined with cycle diagram and cycle spectrum. The results showed that the female mice copulated with C57BL/6J male mice were sterile, and the number of embryos produced in Rev-erb- 偽 -r-female mice was significantly reduced. Rev-erb- 偽 -r-female mouse ovaries had follicles of all levels. The physiological cycle of female mice was 3.22 days longer than that of Rev-erb- 偽 / female mice, and 63.5 days longer than that of Rev-erb- 偽 / female mice. These results suggest that the deletion of Rev-erb- 偽 of the biological clock gene may lead to the prolongation of physiological cycle in female mice, which may be one of the reasons that affect the fertility of female mice. Therefore, the stability of biological rhythm plays an important role in physiological cycle.
