抗炭疽毒素保護(hù)性抗原單克隆抗體的制備及功能分析

【摘要】:目的:原核表達(dá)炭疽桿菌保護(hù)性抗原PA10蛋白,制備單克隆抗體,并進(jìn)行功能分析。方法:人工合成炭疽桿菌保護(hù)性抗原PA10編碼基因并克隆入pUC57載體,酶切鑒定后,將PA10編碼基因插入原核表達(dá)載體pColdⅡ中,測(cè)序驗(yàn)證正確后轉(zhuǎn)化至大腸桿菌BL21(DE3),在IPTG和低溫條件下誘導(dǎo)蛋白表達(dá),SDS-PAGE驗(yàn)證產(chǎn)物;用HiTrap IMAC HP柱純化重組蛋白,Western blot進(jìn)一步驗(yàn)證。以純化獲得PA10重組蛋白為抗原,常規(guī)程序免疫BALB/c小鼠,制備單克隆抗體,并檢測(cè)所制備抗體的中和活性。結(jié)果:獲得的PA10重組蛋白免疫小鼠制備單克隆抗體,最終獲得了4株特異性的單克隆抗體(分別命名為2G8、5A8、7B3、9C9)。經(jīng)體外炭疽毒素保護(hù)試驗(yàn)證實(shí),其中1株單抗(7B3)具有較強(qiáng)的中和活性,其保護(hù)率可高達(dá)96%。結(jié)論:本文成功構(gòu)建并表達(dá)炭疽桿菌保護(hù)性抗原PA10,制備了具有中和活性的抗PA單克隆抗體,為構(gòu)建人源化的抗PA中和抗體奠定基礎(chǔ),從而有望用于炭疽病的治療。
