腸分化細胞黏蛋白O-型糖鏈合成抑制導致MUC2表達減低以及對細菌侵襲易感

【摘要】:目的探討O-型糖鏈合成的抑制對腸分化細胞內(nèi)細菌侵襲數(shù)量和細胞內(nèi)MUC2表達水平的影響。方法對腸分化細胞(HT-29-Gal)用O-型糖鏈抑制劑(benzyl-N-acetyl-α-D-galactosaminide,benzyl-α-GalNAc)處理,采用Real-time PCR和Western blot檢測MUC2 mRNA和蛋白的表達情況。并將HT-29-Gal細胞及benzyl-α-GalNAc處理的HT-29-Gal細胞分別與腸致病性大腸桿菌(enteropathogenic E.coli,EPEC)和腸出血性大腸桿菌(EHEC O157∶H7)37℃孵育2 h,再加入100μg/mL的慶大霉素,殺滅細胞外及粘附于細胞表面的細菌。最后采用系列稀釋克隆計數(shù)法觀察benzyl-α-GalNAc處理的HT-29-Gal細胞對細菌侵襲的影響。結(jié)果 Real-time PCR和Western blot檢測發(fā)現(xiàn)經(jīng)benzyl-α-GalNAc處理的HT-29-Gal細胞MUC2的mRNA和蛋白表達水平明顯降低(P0.05)。侵襲入benzyl-α-GalNAc處理的HT-29-Gal細胞的EPEC和EHEC O157∶H7的數(shù)量較對照細胞顯著增加(P0.05)。結(jié)論抑制HT-29-Gal細胞黏蛋白O-型糖鏈的合成導致侵襲入細胞內(nèi)細菌數(shù)量增加和MUC2的表達降低。
[Abstract]:Objective to investigate the effect of the inhibition of O-type sugar chain synthesis on the number of bacterial invasion and the expression of MUC2 in intestinal differentiation cells.
