慢性間斷性低氧對(duì)大鼠海馬神經(jīng)元及生長相關(guān)蛋白的進(jìn)行性影響

【摘要】:目的觀察慢性間斷性低氧(CIH)過程中大鼠海馬神經(jīng)元的組織結(jié)構(gòu)及生長相關(guān)蛋白(GAP)-43表達(dá)的進(jìn)行性變化。方法成年雄性Sprague-Dawley大鼠40只,隨機(jī)分為對(duì)照組、CIH 1周組、CIH 3周組和CIH 5周組,每組各10只。后三組大鼠在自制缺氧艙內(nèi)每天缺氧8 h,連續(xù)缺氧1周、3周和5周。HE染色后在光學(xué)顯微鏡下觀察海馬神經(jīng)元組織結(jié)構(gòu);免疫組化方法檢測海馬神經(jīng)元GAP-43的表達(dá)。結(jié)果 CIH大鼠海馬出現(xiàn)變性壞死神經(jīng)元,并且隨著缺氧時(shí)間延長,上述改變呈逐漸加重趨勢(shì)。與對(duì)照組比較,CIH組大鼠海馬神經(jīng)元GAP-43陽性表達(dá)增多,CIH后1周出現(xiàn)表達(dá)高峰(P=0.038),CIH后3周表達(dá)逐漸減少(P=0.382),CIH后5周表達(dá)接近正常水平(P=0.860)。結(jié)論隨著缺氧時(shí)間延長,變性壞死神經(jīng)元數(shù)量呈進(jìn)行性增多,而GAP-43的表達(dá)卻與此不一致。
[Abstract]:Aim to observe the progressive changes in the structure of hippocampal neurons and the expression of growth associated protein (GAP-43) in rat hippocampal neurons during chronic intermittent hypoxia (CIH). Methods Forty adult male Sprague-Dawley rats were randomly divided into two groups: the control group (n = 10) and the control group (n = 10). The structure of hippocampal neurons was observed under optical microscope after hypoxia for 8 hours per day in the last three groups, and the expression of GAP-43 in hippocampal neurons was detected by immunohistochemical method. Results denatured and necrotic neurons appeared in the hippocampus of CIH rats. Compared with the control group, the expression of GAP-43 in hippocampal neurons in CIH group was increased. The expression of GAP-43 in hippocampal neurons of CIH group was decreased gradually at 3 weeks after CIH, and the expression of GAP-43 was close to the normal level at 5 weeks after CIH. Conclusion with the prolongation of hypoxia time, the number of denatured and necrotic neurons increased progressively, but the expression of GAP-43 was not consistent with this.
