TNF-α通過(guò)ERK信號(hào)通路刺激骨髓間充質(zhì)干細(xì)胞表達(dá)VCAM-1

【摘要】:本研究目的在于探討TNF-α對(duì)人骨髓間充質(zhì)干細(xì)胞(bone marrow mesenchymal stem cells,BMMSC)血管細(xì)胞黏附分子-1(vascular cell adhesion molecule 1,VCAM-1)表達(dá)的影響及與ERK信號(hào)通路的關(guān)系。應(yīng)用密度梯度離心法結(jié)合貼壁培養(yǎng)法分離人BMMSC,并對(duì)其進(jìn)行表面抗原表達(dá)及誘導(dǎo)分化鑒定。用不同濃度的TNF-α刺激BMMSC,流式細(xì)胞術(shù)檢測(cè)其表面VCAM-1表達(dá)及ERK信號(hào)通路抑制劑U0126對(duì)BMMSC VCAM-1表達(dá)的影響,用Western blot檢測(cè)ERK通路活性的變化。結(jié)果表明,分離培養(yǎng)的BMMSC表達(dá)CD29、CD69、CD44、CD105,不表達(dá)CD34、CD45;BMMSC可誘導(dǎo)分化為成骨細(xì)胞及脂肪細(xì)胞;BMMSC膜上的VCAM-1表達(dá)隨著TNF-α濃度的增加而增加;經(jīng)U0126預(yù)處理后TNF-α刺激的BMMSC膜上的VCAM-1表達(dá)減少;TNF-α增加BMMSC ERK信號(hào)通路的活化程度,U0126抑制TNF-α對(duì)ERK活性的作用。結(jié)論:TNF-α可能通過(guò)ERK信號(hào)通路刺激MSC表達(dá)VCAM-1。
[Abstract]:The aim of this study was to investigate the effect of TNF- 偽 on the expression of vascular adhesion molecule-1 (- 1) and vascular cell adhesion molecule 1 (VCAM-1) in human bone marrow mesenchymal stem cells (BMSCs) and the relationship between TNF- 偽 and ERK signaling pathway. Human BMMSCS were isolated by density gradient centrifugation combined with adherent culture, and their surface antigen expression and differentiation were identified. BMMSCs were stimulated with different concentrations of TNF- 偽. The expression of VCAM-1 on the surface and the effect of U0126, a signal pathway inhibitor of ERK, on the expression of BMMSC VCAM-1 were detected by flow cytometry. The activity of ERK pathway was detected by Western blot. The results showed that BMMSC expressed CD29, CD69, CD44, CD105, and no CD34, CD45, BMMSC could induce differentiation into osteoblasts and adipocytes, and the VCAM-1 expression on BMMSC membrane increased with the increase of TNF- 偽 concentration. After pretreatment with U0126, the expression of VCAM-1 on BMMSC membrane stimulated by TNF- 偽 decreased and TNF- 偽 increased the activation of BMMSC ERK signaling pathway. U0126 inhibited the effect of TNF- 偽 on the activity of ERK. ConclusionTNF- 偽 may stimulate MSC expression of VCAM-1through ERK signaling pathway.
