【摘要】：目的建立尖吻蝮蛇血凝酶(商品名蘇靈,代號(hào)Saculin)多克隆抗體檢測(cè)的間接ELISA法,并應(yīng)用于其臨床免疫學(xué)檢測(cè)。方法免疫家兔獲得多克隆抗體,Western blot鑒定其特異性,通過(guò)比較各種不同條件的檢測(cè)結(jié)果來(lái)確立檢測(cè)尖吻蝮蛇血凝酶多克隆抗體的間接ELISA法的最佳工作條件,并以此為參考檢測(cè)臨床血清。結(jié)果間接ELISA法測(cè)定兔抗血清效價(jià)高達(dá)1∶6 553 600,55例臨床試驗(yàn)受試者(36例為Saculin實(shí)驗(yàn)組,19例為安慰劑組)給藥3周后血清抗體檢測(cè)結(jié)果均為陰性。結(jié)論本研究建立的抗尖吻蝮蛇血凝酶多克隆抗體間接ELISA法能夠用于臨床血清的判定。
[Abstract]:Objective to establish an indirect ELISA method for the detection of polyclonal antibodies against hemocoagulase of Agkistrodon acutus.Methods the rabbit was immunized with polyclonal antibody by Western blot. The best working conditions of indirect ELISA method for the detection of hemagglutinin polyclonal antibody of Agkistrodon acutus were established by comparing the results of different detection conditions.The clinical serum was detected with this reference.Results the titer of rabbit antiserum by indirect ELISA method was as high as 1:6 553,600 and 55 clinical subjects, 36 cases were Saculin experimental group (19 cases were placebo group) 3 weeks after administration, the results of serum antibody detection were all negative.Conclusion the indirect ELISA assay against Agkistrodon acutus hemagglutinin polyclonal antibody developed in this study can be used to determine clinical serum.
