人血小板表面CD36糖蛋白檢測方法的建立及初步應用

【摘要】:個體血小板表面CD36抗原缺乏在隨機輸注時有產生抗-CD36免疫反應的風險,是血小板輸注無效的原因之一。本研究應用流式細胞術檢測杭州地區(qū)單采血小板供者的血小板表面CD36抗原表達情況,并分析個體血小板上CD36缺失表型的頻率。留取獻血者新鮮抗凝血樣,經離心獲取富血小板血漿,洗滌并調整血小板計數至1×106。采用CD36-FITC、CD41-PE單克隆抗體和血小板孵育反應,然后用流式細胞儀檢測和分析血小板表面糖蛋白CD36抗原表達情況。對于血小板表面CD36抗原陰性的標本,進一步篩查其單核細胞表面CD36的表達情況。結果表明:192例無償獻血者篩查出7例血小板表面CD36抗原陰性,CD36缺失型頻率為3.6%,均為Ⅱ型缺失。人群中個體CD36抗原表達強度存在差異,參照CD36幾何平均熒光強度數值大小,59例為低表達,126例為高表達。結論:人群中存在CD36Ⅱ型缺失表型,這些數據將為研究CD36抗原分布提供參考,有助于解決血小板輸注無效問題。
[Abstract]:Individual platelet surface CD36 antigen deficiency is at risk of anti-CD36 immunoreaction during random infusion. In this study, flow cytometry was used to detect the expression of CD36 antigen on platelet surface of single platelet donors in Hangzhou area. The frequency of CD36 deletion phenotype on individual platelets was analyzed. Fresh anticoagulant blood samples were collected from blood donors, platelet-rich plasma was obtained by centrifugation, platelet count was washed and adjusted to 1 脳 106. Monoclonal antibody CD36-FITC+ CD41-PE and platelet incubation reaction were used. Then flow cytometry was used to detect and analyze the expression of glycoprotein CD36 antigen on platelet surface.
