【摘要】：個(gè)體血小板表面CD36抗原缺乏在隨機(jī)輸注時(shí)有產(chǎn)生抗-CD36免疫反應(yīng)的風(fēng)險(xiǎn),是血小板輸注無(wú)效的原因之一。本研究應(yīng)用流式細(xì)胞術(shù)檢測(cè)杭州地區(qū)單采血小板供者的血小板表面CD36抗原表達(dá)情況,并分析個(gè)體血小板上CD36缺失表型的頻率。留取獻(xiàn)血者新鮮抗凝血樣,經(jīng)離心獲取富血小板血漿,洗滌并調(diào)整血小板計(jì)數(shù)至1×106。采用CD36-FITC、CD41-PE單克隆抗體和血小板孵育反應(yīng),然后用流式細(xì)胞儀檢測(cè)和分析血小板表面糖蛋白CD36抗原表達(dá)情況。對(duì)于血小板表面CD36抗原陰性的標(biāo)本,進(jìn)一步篩查其單核細(xì)胞表面CD36的表達(dá)情況。結(jié)果表明:192例無(wú)償獻(xiàn)血者篩查出7例血小板表面CD36抗原陰性,CD36缺失型頻率為3.6%,均為Ⅱ型缺失。人群中個(gè)體CD36抗原表達(dá)強(qiáng)度存在差異,參照CD36幾何平均熒光強(qiáng)度數(shù)值大小,59例為低表達(dá),126例為高表達(dá)。結(jié)論:人群中存在CD36Ⅱ型缺失表型,這些數(shù)據(jù)將為研究CD36抗原分布提供參考,有助于解決血小板輸注無(wú)效問(wèn)題。
[Abstract]:Individual platelet surface CD36 antigen deficiency is at risk of anti-CD36 immunoreaction during random infusion. In this study, flow cytometry was used to detect the expression of CD36 antigen on platelet surface of single platelet donors in Hangzhou area. The frequency of CD36 deletion phenotype on individual platelets was analyzed. Fresh anticoagulant blood samples were collected from blood donors, platelet-rich plasma was obtained by centrifugation, platelet count was washed and adjusted to 1 脳 106. Monoclonal antibody CD36-FITC+ CD41-PE and platelet incubation reaction were used. Then flow cytometry was used to detect and analyze the expression of glycoprotein CD36 antigen on platelet surface.
