建立SD大鼠胚胎-胎仔發(fā)育指標(biāo)的背景數(shù)據(jù)

【摘要】:目的總結(jié)本研究中心的SD大鼠胚胎-胎仔發(fā)育毒性試驗(yàn)各個(gè)指標(biāo)的正常值范圍,建立SD大鼠胚胎-胎仔發(fā)育指標(biāo)的背景數(shù)據(jù),為生殖毒理研究提供參考。方法從本研究中心近期的生殖毒性試驗(yàn)中選取多批SD大鼠胚胎-胎仔發(fā)育毒性試驗(yàn),整理對(duì)照組孕鼠的胚胎發(fā)育和胎仔生長(zhǎng)發(fā)育指標(biāo)數(shù)據(jù),包括孕鼠的窩平均黃體數(shù)、著床數(shù)、平均活胎數(shù)、活胎率、吸收胎率、死胎率,胎仔體重、胎盤重、身長(zhǎng)、尾長(zhǎng)、外觀畸形、內(nèi)臟和骨骼發(fā)育指標(biāo)等,將數(shù)據(jù)進(jìn)行統(tǒng)計(jì),分別計(jì)算各個(gè)指標(biāo)的均數(shù)和標(biāo)準(zhǔn)差。結(jié)果本研究總結(jié)了214只孕鼠和2734只活胎仔的胚胎-胎仔發(fā)育指標(biāo)。本研究所總結(jié)的孕鼠年齡約為50~90日齡,孕鼠妊娠第0天(GD 0)體重為(229±20)g,妊娠第20天(GD 20)體重為(380±34)g,整個(gè)孕期體重增重較大,食量隨孕期增長(zhǎng)而逐漸增加。GD 20安樂(lè)死并實(shí)施解剖,孕鼠窩平均黃體數(shù)為(15.6±2.8)個(gè),窩平均著床數(shù)為(13.6±3.7)個(gè),窩平均活胎數(shù)為(12.8±3.7)只,總吸收胎數(shù)為177(177/2734),早期吸收胎數(shù)為175(175/2734),晚期吸收胎數(shù)為2(2/2734),死胎數(shù)為0(0/2734),活胎率為(93.96±9.54)%,吸收胎率為(6.04±9.54)%,早期吸收胎率為(5.98±9.40)%,晚期吸收胎率為(0.07±0.69)%,死胎率為(0.00±0.00)%。胎仔發(fā)育指標(biāo)中,雌性胎仔體重為(3.751±0.325)g,雄性胎仔體重為(3.942±0.364)g,胎仔胎盤重為(0.558±0.112)g,胎仔身長(zhǎng)(37.17±1.41)mm,尾長(zhǎng)為(13.17±0.79)mm,胎仔性別比為雄性/雌性=1291/1443=0.9;钐プ兄,胎仔外觀畸形率為(0.06±0.64)%;骨骼變異胎仔率為(4.22±13.24)%,骨骼異常胎仔率為(2.27±8.17)%;內(nèi)臟變異胎仔率為(25.81±25.51)%,內(nèi)臟異常胎仔率為(0.88±4.43)%。結(jié)論本中心的SD大鼠胚胎-胎仔發(fā)育毒性試驗(yàn)各個(gè)指標(biāo)的改變均比較穩(wěn)定,胎仔自發(fā)畸形率較低。
[Abstract]:Objective to summarize the range of normal values of SD rat embryo-fetal developmental toxicity test, and to establish the background data of SD rat embryonic fetal development index. Methods A number of batches of SD rat embryo-fetal developmental toxicity tests were selected from the recent reproductive toxicity tests in this research center to collate the data of embryonic development and fetal growth and development of pregnant rats in the control group. Including the average number of corpus luteum, the number of implantation sites, the average number of live births, the rate of live foetus, the rate of absorption, the rate of stillbirth, the body weight, placenta weight, body length, tail length, appearance deformity, visceral and bone development indicators of pregnant mice. Results the embryonic development indexes of 214 pregnant rats and 2734 live foetus were summarized. The gestational age was about 50 ~ 90 days. The body weight of pregnant rats was 229 鹵20g on day 0 of gestation and 380 鹵34g of GD20 on the 20th day of gestation. The body weight of the whole pregnancy increased greatly, and the food intake increased gradually with the increase of pregnancy. The mean number of corpus luteum of pregnant mice was 15.6 鹵2.8), and the mean number of corpus luteum of pregnant mice was 15.6 鹵2.8. The body weight of female foetus was 3.751 鹵0.325 渭 g, that of male fetus was 3.942 鹵0.364 g, that of placenta was 0.558 鹵0.112g, that of foetus was 37.17 鹵1.41g, and that of tail was 13.17 鹵0.79nm. the sex ratio of male to female was 1291r14430.0.The sex ratio of male and female was 1291r14430.9.The body weight of female foetus was 3.75 鹵0.325g, and that of male foetus was 3.942 鹵0.364g, the placenta weight of foetus was 0.558 鹵0.112g. The deformity rate of fetal appearance was 0.06 鹵0.64, the abnormal fetal rate was 2.27 鹵8.17, the abnormal fetal rate was 2.27 鹵8.17, the abnormal fetal rate was 25.81 鹵25.51 and the abnormal visceral fetal rate was 0.88 鹵4.430.Conclusion the developmental toxicity test of SD rat embryo and fetus in this center is of significance. The changes are relatively stable. The rate of spontaneous malformation of fetus was lower.
