腰痹通膠囊對(duì)大鼠自體髓核移植模型中背根節(jié)組織形態(tài)學(xué)和磷酸化p38MAPK表達(dá)的影響

【摘要】:目的:通過(guò)腰痹通膠囊對(duì)大鼠自體髓核移植模型中背根節(jié)組織學(xué)和磷酸化p38MAPK表達(dá)影響的研究,探討腰痹通膠囊對(duì)治療腰椎間盤(pán)突出癥的作用機(jī)制,對(duì)于今后中藥的有效成分的分析、藥物的提純及制劑工藝和臨床藥理作用的深入研究將起到肯定作用。方法:將SD大鼠隨機(jī)分為空白組、模型組以及腰痹通膠囊組,其中后兩組建立自體髓核移植模型,模型組給予生理鹽水,腰痹通膠囊組連續(xù)給藥30天。觀察記錄各組大鼠背根神經(jīng)節(jié)的組織病理學(xué)、組織形態(tài)學(xué)及對(duì)機(jī)械刺激壓痛閾值、磷酸化p38MAPK及其他炎癥因子表達(dá)水平,將所得數(shù)據(jù)進(jìn)行統(tǒng)計(jì)分析。結(jié)果:模型組大鼠背根神經(jīng)節(jié)形態(tài)學(xué)表現(xiàn)核膜出現(xiàn)邊界不清晰,且在細(xì)胞漿中出現(xiàn)空泡樣的組織,細(xì)胞漿中的尼氏小體分布較為紊亂,核仁染色變淡甚至消失等形態(tài)學(xué)表現(xiàn),但是腰痹通膠囊組實(shí)驗(yàn)大鼠中的背根神經(jīng)節(jié)的組織形態(tài)學(xué)發(fā)生了顯著的改善,核膜邊界由不清晰逐漸變得比較清晰,并且胞漿中充滿(mǎn)細(xì)顆粒狀物(尼氏小體),尼氏小體的分布也由紊亂狀態(tài)變?yōu)橄鄬?duì)比較均勻的狀態(tài),核仁也由染色變淡甚至消失變?yōu)楦忧逦黠@;與空白組相比,模型組與腰痹通膠囊組的初始?jí)和撮撝挡町悷o(wú)統(tǒng)計(jì)學(xué)意義(P0.05);與空白組相比,模型組的最終壓痛閾值顯著降低,差異具有統(tǒng)計(jì)學(xué)意義(P0.01);與模型組相比,腰痹通膠囊組的最終壓痛閾值顯著降升高,差異具有統(tǒng)計(jì)學(xué)意義(P0.05);模型組中的磷酸化p38MAPK的蛋白表達(dá)水平顯著高于空白組;腰痹通膠囊組中的磷酸化p38MAPK的蛋白表達(dá)水平明顯低于模型組。結(jié)論:腰痹通膠囊對(duì)大鼠自體髓核移植模型的癥狀于背根神經(jīng)節(jié)組織的形態(tài)學(xué)變化能夠發(fā)揮改善作用,腰痹通膠囊可以降低腫瘤壞死因子-α與白介素-1的表達(dá),其作用機(jī)制可能與影響磷酸化p38MAPK的表達(dá)水平有關(guān),進(jìn)而抑制炎癥因子在機(jī)體中的表達(dá),改善不良癥狀。腰痹通膠囊作為一種治療腰椎間盤(pán)突出癥尤其是非壓迫因素引發(fā)的背根神經(jīng)性疼痛疾病的藥物具有良好的臨床效果,具有良好的臨床應(yīng)用前景。
[Abstract]:Objective: to study the effect of Yaobitong capsule on the expression of phosphorylated p38 MAPK in dorsal root ganglion (DRG) of rats in order to explore the mechanism of Yaobitong capsule in the treatment of lumbar disc herniation. For the analysis of the effective components of traditional Chinese medicine in the future, the purification of the drug and the further study of the preparation technology and clinical pharmacological action will play a positive role. Methods: SD rats were randomly divided into blank group, model group and Yaobitong capsule group. The model of autologous nucleus pulposus transplantation was established in the latter two groups. The rats in the model group were given normal saline and Yaobitong capsule for 30 days. The histopathology, histomorphology and the threshold of tenderness to mechanical stimulation were observed and recorded in the dorsal root ganglion of rats in each group. The expression levels of phosphorylated p38 MAPK and other inflammatory factors were analyzed statistically. Results: the morphological features of the dorsal root ganglion in the model group showed that the nuclear membrane was not clear and vacuolated tissue appeared in the cytoplasm. The distribution of Nissl corpuscles in the cytoplasm was disordered, the nucleolus staining became light or even disappeared, but the histomorphology of the dorsal root ganglion in the experimental rats of Yaobitong capsule group was significantly improved. The boundary of the nuclear membrane gradually became clearer and the cytoplasm was filled with fine particles. (the distribution of the Nissl bodies changed from a disordered state to a relatively uniform state. Compared with the blank group, the initial tenderness threshold of model group and Yaobitong capsule group was not significantly different (P 0.05), and the final tenderness threshold of model group was significantly lower than that of blank group. Compared with the model group, the final tenderness threshold of Yaobitong capsule group decreased significantly, and the difference was statistically significant (P 0.05), the expression of phosphorylated p38 MAPK protein in the model group was significantly higher than that in the blank group. The protein expression level of phosphorylated p38 MAPK in Yaobitong capsule group was significantly lower than that in model group. Conclusion: Yaobitong capsule can improve the symptoms of autologous nucleus pulposus transplantation model in rats and improve the morphological changes of dorsal root ganglion tissue. Yaobitong capsule can reduce the expression of tumor necrosis factor- 偽 and interleukin-1, its mechanism may be related to the effect of phosphorylation of p38 MAPK expression level, and then inhibit the expression of inflammatory factors in the body. As a drug to treat lumbar disc herniation, especially non-compression factors, Yaobitong capsule has a good clinical effect and has a good clinical application prospect.
