【摘要】:【目的】探討超順磁性氧化鐵(SPIO)標(biāo)記大鼠脂肪間充質(zhì)干細(xì)胞(RADSC)后,其成骨樣分化細(xì)胞中鐵調(diào)節(jié)蛋白1(IRP1)基因及蛋白表達(dá)的變化!痉椒ā勘緦嶒灢捎棉D(zhuǎn)染劑多聚賴氨酸(PLL)介導(dǎo)SPIO標(biāo)記RADSC,然后將其誘導(dǎo)為成骨樣分化細(xì)胞,分別在誘導(dǎo)后0、7、14、21 d進(jìn)行RT-PCR及Western blot檢測其IRP1基因及蛋白的表達(dá)!窘Y(jié)果】成骨樣分化細(xì)胞IRP1 mRNA表達(dá)水平在SPIO標(biāo)記與未標(biāo)記組之間差異均無統(tǒng)計學(xué)顯著性意義(F=1.571,P=0.239);IRP1蛋白表達(dá)水平在標(biāo)記與未標(biāo)記組之間差異均無統(tǒng)計學(xué)顯著性意義(F=0.547,P=0.500)!窘Y(jié)論】SPIO標(biāo)記并未對成骨樣分化細(xì)胞的IRP1基因及IRP1蛋白表達(dá)產(chǎn)生明顯影響。
[Abstract]:[objective] to investigate the labeling of rat adipose mesenchymal stem cells (RADSCs) by superparamagnetic iron oxide (SPIO). The changes of iron regulatory protein 1 (IRP1) gene and protein expression in osteoblast-like differentiation cells. [methods] SPIO labeled RADSC was mediated by polylysine transducer (PLL). Then they were induced into osteoblast-like differentiation cells, respectively. RT-PCR and Western blot were used to detect the expression of IRP1 gene and protein in 21 days. [results] Osteoblast-like differentiation cell IRP1. There was no significant difference in the expression of mRNA between SPIO labeled and unlabeled groups (P < 0.05). FN 1.571. Pu 0.239; There was no significant difference in the expression of IRP1 protein between labeled and unlabeled groups. [conclusion] SPIO labeling has no significant effect on the expression of IRP1 gene and IRP1 protein in osteoblast-like differentiation cells.
