【摘要】:背景:寡核苷酸結(jié)合摺疊區(qū)1基因(oligonucleotide/oligosaccharide binding fold containing 1 OBFC1)與酵母的Stn1基因具有同源性,并且是直接參與端粒生物功能調(diào)節(jié)的重要基因之一[1,2]。迄今為止,國(guó)內(nèi)外對(duì)OBFC1基因多態(tài)性與血脂的相關(guān)性研究甚少,為了了解該基因與血脂的關(guān)系,我們進(jìn)行了相關(guān)研究。目的:探討OBFC1 rs4387287多態(tài)性與血脂水平的關(guān)系,了解該基因?qū)V西紅水河流域長(zhǎng)壽人群的影響。方法:將2262名研究對(duì)象分為四組:長(zhǎng)壽組為廣西紅水河流域的長(zhǎng)壽老人,年齡90歲,平均年齡93.27±3.01,共496人;長(zhǎng)壽子女組為廣西紅水河流域的長(zhǎng)壽老人的子女,年齡40-75歲,平均年齡59.84±7.81,共723人;當(dāng)?shù)貙?duì)照組為廣西紅水河流域的非長(zhǎng)壽家系老人,年齡40-75歲,平均年齡63.42±7.77,共611人;平果對(duì)照組為平果地區(qū)的非長(zhǎng)壽家系老人,年齡40-75歲,平均年齡60.58±7.99,共432人;所選人群均為壯族(參考身份證件)。本次研究主要對(duì)身高、體重、血壓、血脂、血糖等指標(biāo)進(jìn)行測(cè)量,應(yīng)用改良多重酶連接反應(yīng)(improved multiplex ligation detection reaction,i MLDR)技術(shù)對(duì)OBFC1 rs4387287位點(diǎn)進(jìn)行分型,從而了解該基因多態(tài)性對(duì)血脂水平的影響。結(jié)果:1.各臨床資料(血脂、血糖和血壓)的組間比較長(zhǎng)壽組的TG、HDL-C水平低于長(zhǎng)壽子女組和當(dāng)?shù)貙?duì)照組(均P0.05),但SBP、DBP卻出現(xiàn)相反趨勢(shì)。平果對(duì)照組的tc、ldl-c、sbp、血糖水平低于長(zhǎng)壽子女組和當(dāng)?shù)貙?duì)照組(均p0.05)。2.obfc1rs4387287基因型及等位基因頻率分布情況在總?cè)巳褐?基因型以cc基因型較為多見,頻率為71.3%;ca基因型的頻率為24.5%;aa基因型的頻率為4.2%;c等位基因的頻率為83.6%;a等位基因頻率為16.4%。a等位基因在長(zhǎng)壽組的頻率為16.2%;在長(zhǎng)壽子女組的頻率為15.8%;在當(dāng)?shù)貙?duì)照組的頻率為15.3%;在平果對(duì)照組的頻率為19.2%,其中a等位基因在平果對(duì)照組的頻率高于其余三組(均p0.05)。在當(dāng)?shù)貙?duì)照組中,男性cc基因型頻率高于同組女性,a等位基因頻率低于同組女性(均p0.05)。3.obfc1rs4387287基因型及等位基因?qū)θ巳貉降挠绊懺谄焦麑?duì)照組中,cc基因型的tc水平高于非cc基因型(p=0.014)。性別分層后,在總體男性人群中,cc基因型的hdl-c水平低于非cc基因型(p=0.016);在平果對(duì)照組男性中,cc基因型的tc、tg水平高于非cc基因型(p分別為0.008、0.009);在長(zhǎng)壽子女組及當(dāng)?shù)貙?duì)照組的男性群體中,cc基因型的tc水平低于非cc基因型(p=0.028)。血脂分層后,在正常血脂亞組中,tg水平高于非cc基因型(p=0.001),hdl-c水平低于非cc基因型(p=0.041);在長(zhǎng)壽子女組的正常血脂亞組中,cc基因型tg、hdl-c水平低于非cc基因型(p分別為0.014、0.037);在異常血脂亞組中均未發(fā)現(xiàn)統(tǒng)計(jì)學(xué)差異。bmi分層后,在正常bmi亞組內(nèi),長(zhǎng)壽組的cc基因型的hdl-c水平低于非cc基因型(p=0.023);在平果對(duì)照組中,cc基因型的tc、ldl-c水平高于非cc基因型(p分別為0.010、0.013)。4.obfc1rs4387287基因型、年齡等因素與血脂水平的回歸分析tc、tg、ldl-c水平主要與bmi、性別、年齡、舒張壓呈現(xiàn)正相關(guān);hdl-c水平主要與bmi、性別、年齡、體重、舒張壓呈現(xiàn)負(fù)相關(guān)。性別分層后,在長(zhǎng)壽子女組及當(dāng)?shù)貙?duì)照組的男性人群中,OBFC1 rs4387287基因型(AA)與TC水平呈現(xiàn)正相關(guān);在平果對(duì)照組男性中,OBFC1 rs4387287基因型(AA)與TC、TG水平呈現(xiàn)負(fù)相關(guān)。結(jié)論:1.OBFC1 rs4387287多態(tài)性與TC、TG水平相關(guān),其A等位基因可能與平果地區(qū)人群較低的血脂水平有關(guān)。2.OBFC1 rs4387287多態(tài)性的A等位基因在平果對(duì)照組人群中有富集現(xiàn)象,與該地男性群體較低的TC、TG水平有關(guān);但其對(duì)廣西紅水河流域人群的血脂水平影響不明顯。
[Abstract]:Background: the oligonucleotide binding fold (oligonucleotide/oligosaccharide binding fold gene 1 containing 1 OBFC1) with homology with the Stn1 gene of the yeast, and is directly involved in the regulation of telomere biological function is one of the important genes of [1,2]. so far, the domestic and foreign research on the correlation between OBFC1 gene polymorphism and blood lipids is poorly understood, in order to understand the relationship between the gene and blood lipids. We carried out relevant research. Objective: To investigate the relationship between OBFC1 rs4387287 polymorphism and serum lipid levels, to understand the effect of this gene on Guangxi Red River Basin longevity population. The clinical results: 1. the data (blood lipids, blood glucose and blood pressure) between the two groups in longevity Group TG, HDL-C level is lower than the longevity of children group and local controls (all P0.05), but SBP, DBP shows the opposite trend. Pingguo control group TC, LDL-C, SBP, blood glucose level is lower than the longevity of the children group And the control group (P0.05).2.obfc1rs4387287 genotype distribution and allele frequency in the general population, the genotype CC genotype is more common, the frequency is 71.3%; the frequency of Ca genotype was 24.5%; the frequency of AA genotype was 4.2%; the frequency of allele C was 83.6%; a the allele frequency of 16.4%.a allele in the longevity Group was 16.2% in the children group; longevity frequency was 15.8% in the control group; the local frequency is 15.3% in the control group; Pingguo frequency was 19.2%, the A allele frequency in Pingguo control group than the other three groups (P0.05).
