【摘要】：目的采用Ⅰ型膠原酶原位灌注消化法原位獲取兔頸靜脈的內(nèi)皮細(xì)胞,構(gòu)建兔頸靜脈的內(nèi)皮剝脫和血栓形成模型。方法新西蘭大白兔12只,雌雄不拘,隨機(jī)分入抗凝組和對(duì)照組。①分離兔右側(cè)頸靜脈,保留對(duì)側(cè)頸靜脈,采用改進(jìn)的Ⅰ型膠原酶原位灌注消化法分離兔頸靜脈內(nèi)皮細(xì)胞,體外培養(yǎng),并進(jìn)行血管內(nèi)皮細(xì)胞鑒定。②抗凝組兔腹部皮下注射低分子肝素鈉,對(duì)照組不給予抗凝血藥物,7d后處死動(dòng)物,檢查右頸靜脈的通暢情況。結(jié)果抗凝組中,1只兔在術(shù)后第3天出現(xiàn)口腔和鼻腔出血,糞便帶血,手術(shù)切口出血而死亡,其右側(cè)頸靜脈通暢,頸部切口內(nèi)有明顯血腫。存活5只:頸靜脈通暢2只;頸靜脈內(nèi)少量血栓形成1只,管腔輕度狹窄,管腔殘余內(nèi)徑為對(duì)側(cè)頸靜脈的80%;頸靜脈內(nèi)中等程度血栓形成1只,管腔中度狹窄,管腔殘余內(nèi)徑為對(duì)側(cè)頸靜脈的50%;兔頸靜脈內(nèi)血栓形成、管腔完全堵塞1只。對(duì)照組中,術(shù)后第2天開始出現(xiàn)球結(jié)膜水腫1只,肢體和軀干運(yùn)動(dòng)減少,術(shù)后第4天死亡,其右側(cè)頸靜脈內(nèi)血栓形成,管腔完全堵塞,血栓向下延伸到上腔靜脈,造成上腔靜脈管腔堵塞;右側(cè)頸靜脈內(nèi)血栓形成、管腔完全堵塞5只,均存活。結(jié)論Ⅰ型膠原酶原位灌注消化法構(gòu)建兔頸靜脈去內(nèi)皮和血栓形成模型具有一定的可行性,可為研究頸靜脈血管去內(nèi)皮后如何預(yù)防血栓形成的藥物學(xué)方法和血栓形成以后的治療手段提供實(shí)驗(yàn)對(duì)象。
[Abstract]:Objective to obtain endothelial cells from rabbit jugular vein in situ by type I collagenase in situ perfusion method, and to construct the model of endothelial denudation and thrombosis in the jugular vein of rabbits. Methods 12 New Zealand white rabbits were randomly divided into the anticoagulant group and the control group. First, the right jugular vein was isolated from the right side of the jugular vein, and the contralateral jugular vein was reserved. The rabbit carotid vein endothelial cells were isolated by improved type I collagenase perfusion in situ, and cultured in vitro, and the endothelial cells were identified. The low molecular weight heparin sodium was injected subcutaneously into the abdomen of the anticoagulant group, the control group did not give anticoagulant drugs, and the animals were killed after 7d, and the patency of the right jugular vein was checked. Results in the anticoagulant group, 1 rabbits developed oral and nasal bleeding on the third day after operation. The blood was excreted from the stool and died after operation incision bleeding. The right jugular vein was unobstructed and there was obvious hematoma in the neck incision. Only 5 survived: jugular venous patency 2; jugular vein and small thrombosis in 1, mild stenosis, residual lumen diameter of jugular vein 80%; jugular vein thrombosis in moderate 1, moderate stenosis lumen diameter, lumen residual contralateral jugular vein 50%; rabbit jugular vein thrombosis the formation, lumen completely blocked 1. In the control group, second days postoperatively chemosis 1, limbs and trunk movements reduced, died fourth days after the operation, the right jugular vein thrombosis, lumen completely blocked, thrombus extending down into the superior vena cava, resulting in superior vena cava lumen; right jugular vein thrombosis, lumen completely block 5, survived. Conclusion typeI in situ collagenase perfusion digestion method to construct rabbit jugular vein endothelial and thrombosis model has certain feasibility to study endothelial jugular vein after how to prevent thrombosis after pharmacological method and formation treatment for experiment.
