α-烯醇化酶在慢性阻塞性肺疾病合并肺腺癌患者肋間肌水平的研究

【摘要】:目的通過(guò)測(cè)定α-烯醇化酶及其mRNA在肺腺癌、慢性阻塞性肺疾病(COPD)合并肺腺癌患者肋間肌中的表達(dá)情況,探討α-烯醇化酶在COPD中的調(diào)控情況。方法選取2012年4—10月就診于昆明醫(yī)科大學(xué)第一附屬醫(yī)院胸外科需行開(kāi)胸腫瘤切除術(shù)的住院患者28例。根據(jù)術(shù)后腫瘤組織病理類(lèi)型和中華醫(yī)學(xué)會(huì)呼吸學(xué)分會(huì)慢性阻塞性肺疾病學(xué)組頒布的《慢性阻塞性肺疾病診療指南》(2008年修訂版)中的診斷標(biāo)準(zhǔn)分為肺腺癌組12例和COPD合并肺腺癌組16例。采用實(shí)時(shí)定量熒光PCR(RT-PCR)測(cè)定α-烯醇化酶mRNA表達(dá)水平,采用ELISA法測(cè)定α-烯醇化酶蛋白水平。結(jié)果 COPD合并肺腺癌組α-烯醇化酶mRNA表達(dá)水平為(3.53±0.84),高于肺腺癌組的(2.36±0.63)(t=10.53,P0.01)。COPD合并肺腺癌組α-烯醇化酶蛋白水平為(1 175.51±219.16)ng/L,高于肺腺癌組的(1 047.29±120.83)ng/L(t=3.569,P0.01)。COPD合并肺腺癌患者肋間肌α-烯醇化酶mRNA表達(dá)水平與性別、年齡、第1秒用力呼氣末容積占用力肺活量百分比(FEV1/FVC)、氧分壓(PO2)、二氧化碳分壓(PaCO2)無(wú)直線(xiàn)相關(guān)關(guān)系(r=0.670、0.545、0.498、0.658、0.420,P0.05),與第1秒用力呼氣末容積占預(yù)計(jì)值百分比(FEV1/Pred)呈負(fù)相關(guān)(r=-0.713,P0.05)。COPD合并肺腺癌患者肋間肌α-烯醇化酶蛋白水平與性別、年齡、FEV1/FVC、PO2、PaCO2無(wú)直線(xiàn)相關(guān)關(guān)系(r=0.544、0.608、0.435、0.590、0.498,P0.05),與FEV1/Pred呈負(fù)相關(guān)(r=-0.819,P0.05)。結(jié)論 COPD可能通過(guò)調(diào)控α-烯醇化酶上游基因使肋間肌α-烯醇化酶mRNA表達(dá)水平及蛋白表達(dá)異常,從而影響患者肋間肌功能狀態(tài)。
[Abstract]:Aim to determine the expression of 偽-enolase and its mRNA in intercostal muscles of patients with lung adenocarcinoma, chronic obstructive pulmonary disease (COPD) and lung adenocarcinoma, and to explore the regulation of 偽-enolase in COPD. Methods from April to October 2012, 28 inpatients who needed thoracotomy in the first affiliated Hospital of Kunming Medical University were selected. According to the pathological types of postoperative tumor tissue and the diagnostic criteria of chronic obstructive pulmonary disease (COPD) issued by the respiratory branch of Chinese Medical Association (revised 2008 edition), the patients were divided into two groups: lung adenocarcinoma group (n = 12) and chronic obstructive pulmonary disease (COPD) group (revised edition, 2008). There were 16 cases of COPD complicated with lung adenocarcinoma. The expression level of 偽-enolase mRNA was measured by real-time quantitative fluorescence PCR (RT-PCR), and the level of 偽-enolase protein was determined by ELISA method. Results the expression level of 偽-enolase mRNA in COPD with lung adenocarcinoma group was (3.53 鹵0.84), which was higher than that in lung adenocarcinoma group (2.36 鹵0.63) (t = 10.53, P < 0.01).
