【摘要】：目的:探索肺血栓栓塞癥(pulmonary thromboembolism,PTE)大鼠肺組織不規(guī)則趨化因子(Fractalkine,FKN)的變化及姜黃素(curcumin,CUR)的干預(yù)作用。方法:采用自體血栓法復(fù)制PTE動(dòng)物模型,96只SD大鼠隨機(jī)分6組:空白組組、假手術(shù)組、模型組、CUR低劑量組、CUR中劑量組、CUR高劑量組,栓塞后4h、72h,分別檢測血清TNF-α、IL-1β、IL-8、FKN,熒光定量PCR檢測肺組織TNF-α、IL-8、FKN的mRNA水平,肺病理HE染色,免疫組化染色檢測CX3CL1及CX3CR1陽性細(xì)胞數(shù)。結(jié)果:栓塞后4h血清ELISA的FKN比較:空白組顯著小于模型組(P0.01),72h后空白組、假手術(shù)組、CUR高劑量組均顯著小于模型組(P0.01);栓塞后4h、72h IL-8比較:空白組、假手術(shù)組、CUR低劑量組、CUR中劑量組顯著小于模型組(P0.05);栓塞后72h血清ELISA的TNF-α比較:空白組、假手術(shù)組、CUR高劑量組均顯著小于模型組(P0.05);栓塞后72h各組血清ELISA的IL-1β均顯著小于模型組(P0.05)。各組栓塞后4h和72h肺組織FKN、TNF-α、IL-1β的mRNA均顯著小于模型組(P0.01)。栓塞后72h大鼠肺HE染色呈肺泡壁血管高度擴(kuò)張、充血,個(gè)別大鼠光鏡下查見出血性梗死灶,應(yīng)用CUR后肺組織充血程度明顯減輕,出現(xiàn)梗死的大鼠數(shù)量也有降低的趨勢。免疫組化發(fā)現(xiàn)栓塞后4h,空白組、假手術(shù)組、CUR高劑量組的CX3CL1陽性細(xì)胞計(jì)數(shù)均顯著小于模型組(P0.01),空白組、假手術(shù)組、CUR中劑量組、CUR高劑量組的CX3CR1陽性細(xì)胞計(jì)數(shù)顯著小于模型組(P0.01)。栓塞后72h,空白組、假手術(shù)組、CUR低劑量組、CUR中劑量組、CUR高劑量組的CX3CL1陽性細(xì)胞計(jì)數(shù)均顯著小于模型組(P0.05),空白組、假手術(shù)組的CX3CR1陽性細(xì)胞計(jì)數(shù)顯著小于模型組(P0.01)。結(jié)論:PTE大鼠存在炎性因子,CUR能抑制PTE大鼠肺TNF-α、IL-1β、IL-8、CX3CL1及CX3CR1的表達(dá),可能是CUR減輕PTE大鼠肺損傷的機(jī)制之一。
[Abstract]:Aim: to investigate the changes of irregular chemokine (Fractalkine,FKN) in pulmonary tissue and the intervention of curcumin (curcumin,CUR) in pulmonary thromboembolism (pulmonary thromboembolism,PTE) rats. Methods: PTE animal model was established by autologous thrombosis. 96 SD rats were randomly divided into 6 groups: blank group, sham operation group, model group, CUR low dose group, CUR medium dose group, CUR high dose group, 4 h after embolization and 72 h after embolization. Serum TNF- 偽, IL-1 尾 and IL-8,FKN, fluorescence quantitative PCR were used to detect the mRNA levels of TNF- 偽 and IL-8,FKN in lung tissue, HE staining in lung pathology, and CX3CL1 and CX3CR1 positive cells in lung tissue by immunohistochemical staining. Results: the comparison of FKN of serum ELISA 4 hours after embolization: the blank group was significantly smaller than the model group (P0.01), the blank group after 72 hours, the sham operation group, the high dose CUR group were significantly smaller than the model group (P0.01). Comparison of IL-8 at 72h after embolization: blank group, sham operation group, CUR low dose group, CUR medium dose group were significantly lower than model group (P0.05); Comparison of TNF- 偽 of serum ELISA at 72 hours after embolization: the IL-1 尾 of serum ELISA in blank group, sham operation group and high dose group of CUR were significantly lower than that in model group (P0.05), and IL-1 尾 in serum of 72 h after embolization was significantly lower than that in model group (P0.05). The mRNA of FKN,TNF- 偽 and IL-1 尾 in lung tissue of each group was significantly lower than that of model group at 4h and 72h after embolization (P0.01). At 72 hours after embolization, the pulmonary HE staining showed high dilation of alveolar wall vessels and congestion. The hemorrhagic infarct was found in some rats under light microscope. The degree of congestion in lung tissue was obviously reduced after the application of CUR, and the number of rats with infarction also decreased. Immunohistochemistry showed that the number of CX3CL1 positive cells in the blank group, sham operation group, high dose CUR group was significantly lower than that in the model group (P0.01), blank group, sham-operation group, CUR medium dose group, the number of CX3CL1 positive cells was significantly lower than that in the model group (P0.01). The number of CX3CR1 positive cells in CUR group was significantly lower than that in model group (P0.01). At 72 hours after embolization, the counts of CX3CL1 positive cells in blank group, sham operation group, CUR low dose group, CUR medium dose group and CUR high dose group were significantly lower than those in model group (P0.05). The number of CX3CR1 positive cells in sham-operated group was significantly lower than that in model group (P0.01). Conclusion: there are inflammatory factors in PTE rats, and CUR can inhibit the expression of TNF- 偽, IL-1 尾, IL-8,CX3CL1 and CX3CR1 in the lung of PTE rats, which may be one of the mechanisms that CUR attenuates the lung injury of PTE rats.
