肌球蛋白輕鏈激酶對(duì)哮喘小鼠氣道炎癥及肺功能的影響

【摘要】:目的研究肌球蛋白輕鏈激酶(MLCK)在哮喘小鼠氣道中的表達(dá)及其對(duì)哮喘氣道炎癥和肺功能的影響。方法將20只雌性BALB/c小鼠隨機(jī)分為對(duì)照組(n=10)及哮喘組(n=10),利用雞卵白蛋白(OVA)建立小鼠哮喘模型。光學(xué)顯微鏡下計(jì)數(shù)血清中白細(xì)胞及嗜酸性粒細(xì)胞總數(shù);ELISA方法檢測(cè)肺泡灌洗液中炎性介質(zhì)IL-4、IFN-γ水平;Western blotting法檢測(cè)小鼠肺組織M LCK的表達(dá);免疫實(shí)時(shí)熒光定量(q PCR)檢測(cè)M LCK在基因水平的表達(dá)。結(jié)果肺功能檢測(cè)示哮喘組小鼠最大呼氣流量峰值(PEF)、第0.4秒用力呼氣容積占用力肺活量比值(FEV0.4/FVC)均低于對(duì)照組(P均0.001);哮喘組小鼠白細(xì)胞及嗜酸性粒細(xì)胞計(jì)數(shù)均高于對(duì)照組(P均0.001);ELISA結(jié)果示哮喘組小鼠BALF中炎性介質(zhì)IL-4濃度較對(duì)照組明顯升高(P0.001),而IFN-γ濃度較對(duì)照組降低(P0.001);哮喘組小鼠肺組織MLCK表達(dá)量明顯升高(P=0.006);與對(duì)照組相比,哮喘組MLCK在基因水平上的表達(dá)升高(P0.001)。結(jié)論 MLCK可能參與哮喘氣道炎癥反應(yīng)并影響肺功能。
[Abstract]:Objective to study the expression of myosin light streptokinase (MLCK) in the airway of asthmatic mice and its effect on airway inflammation and pulmonary function. Methods Twenty female BALB/c mice were randomly divided into two groups: control group (n = 10) and asthmatic group (n = 10). The asthmatic model of mice was established by using chicken ovalbumin (OVA). The total number of leukocytes and eosinophilic granulocytes in serum were counted under optical microscope. The levels of inflammatory mediators IL-4,IFN- 緯 in alveolar lavage fluid were detected by Elisa and the expression of M LCK in lung tissue of mice was detected by Western blotting. The expression of M LCK at gene level was detected by immunofluorescence quantitative (q PCR). Results Pulmonary function test showed that the ratio of forced expiratory volume occupying capacity (FEV0.4/FVC) of peak expiratory flow (PEF),) in asthmatic group was lower than that of control group (P < 0.001), and the counts of leukocytes and eosinophils in asthmatic group were higher than those in control group (P < 0.05). The results of Elisa showed that the concentration of inflammatory mediators IL-4 in BALF of asthmatic group was significantly higher than that of control group (P0. 001), but the concentration of IFN- 緯 was lower than that of control group (P0. 001), and the expression of MLCK in lung tissue of asthmatic group was significantly higher than that of control group (P0. 006). The expression of MLCK in asthma group was increased at the gene level (P0. 001). Conclusion MLCK may be involved in airway inflammation and affect lung function in asthma.
