apelin-APJ在慢性阻塞性肺疾病大鼠肺組織中的表達(dá)及其意義

【摘要】:目的:檢測(cè)慢性阻塞性肺疾病(COPD)大鼠肺組織中apelin及其受體APJ的mRNA和蛋白表達(dá),探討apelin-APJ系統(tǒng)在COPD發(fā)生、發(fā)展中的作用。方法:煙霧暴露法制造COPD大鼠模型。實(shí)驗(yàn)設(shè)對(duì)照組和COPD組。COPD組大鼠每日2次在熏煙箱內(nèi)煙霧暴露,每次連續(xù)被動(dòng)吸煙20支(1 h),持續(xù)4個(gè)月。qRT-PCR測(cè)定大鼠肺組織中apelin mRNA及APJ mRNA表達(dá)。免疫組化法檢測(cè)大鼠肺組織apelin和APJ蛋白表達(dá)。結(jié)果:COPD組apelin、APJ mRNA相對(duì)表達(dá)量顯著低于對(duì)照組(P0.01)。大鼠肺組織apelin、APJ mRNA表達(dá)與RV/(LV+S)均呈負(fù)相關(guān)(r=-0.454、-0.448,均P0.05);與FEV0.3/FVC呈正相關(guān)(r=0.529、0.475,均P0.05)。apelin、APJ蛋白主要表達(dá)在支氣管肺組織上皮細(xì)胞、肺泡巨噬細(xì)胞、血管內(nèi)皮細(xì)胞及部分肺泡壁細(xì)胞的細(xì)胞膜和細(xì)胞漿,COPD組及對(duì)照組apelin、APJ蛋白均呈陽(yáng)性表達(dá),半定量檢測(cè)COPD組表達(dá)低于對(duì)照組(P0.05)。結(jié)論:apelin及其受體APJ的mRNA和蛋白在COPD大鼠肺組織表達(dá)均下降,可能是導(dǎo)致大鼠COPD肺動(dòng)脈高壓、肺心病發(fā)生發(fā)展的重要因素。apelin-APJ系統(tǒng)可能成為探索防治COPD肺動(dòng)脈高壓、右心室肥大的新靶點(diǎn)。
[Abstract]:Aim: to detect the expression of mRNA and protein of apelin and its receptor APJ in lung tissue of (COPD) rats with chronic obstructive pulmonary disease (COPD) and to explore the role of apelin-APJ system in the pathogenesis and development of COPD. Methods: COPD rat model was established by smoke exposure. The rats in the control group and the COPD group were exposed to smoke in the smoke box twice a day, and the expression of apelin mRNA and APJ mRNA in the lung tissue of the rats was determined by qRT-PCR, which lasted for 4 months (1 h), lasted 4 months). The expression of apelin and APJ in rat lung tissue was detected by immunohistochemical method. Results the relative expression of APJ mRNA in apelinia was significantly lower than that in control group (P0.01). The expression of APJ mRNA in rat lung tissue was negatively correlated with RV/ (LV S) (r = 0.4544-0.448, P 0.05), and positively correlated with FEV0.3/FVC (r = 0.529, P < 0.05, all P 0.05). The expression of APJ protein was mainly expressed in bronchopulmonary epithelial cells and alveolar macrophages. The expression of APJ protein in the plasma membrane and cytoplasm of vascular endothelial cells and some alveolar parietal cells was positive in COPD group and control group. The expression of APJ in COPD group was lower than that in control group (P0.05). Conclusion the decreased expression of mRNA and protein in lung tissue of rats with COPD may be an important factor in the pathogenesis and development of COPD pulmonary hypertension. The apelin-APJ system may be an important factor to explore the prevention and treatment of COPD pulmonary hypertension. A new target for right ventricular hypertrophy.
