HIF-1α在間歇低氧合并肺氣腫大鼠肝細(xì)胞凋亡中的機(jī)制研究

【摘要】:目的探討間歇低氧(IH)合并肺氣腫對(duì)大鼠肝臟低氧誘導(dǎo)因子-1α(HIF-1α)、Bax、Bcl-2表達(dá)的影響及HIF-1α在肝細(xì)胞凋亡中的作用機(jī)制。方法將60只大鼠按隨機(jī)數(shù)字表法均分為4組:正常對(duì)照組,正常飼養(yǎng);IH組,每天9:00—17:00于低氧艙內(nèi)交替通入30 s氮?dú)夂?0 s空氣,8 h/d;肺氣腫組,每天8:00和18:00于熏箱內(nèi)熏煙各30 min;IH合并肺氣腫組,上述IH組和肺氣腫組方法的聯(lián)合。暴露14周后,處死大鼠,采用熒光實(shí)時(shí)定量(qRT)-PCR法測(cè)定肝組織中HIF-1α、Bax、Bcl-2表達(dá)水平。結(jié)果 IH合并肺氣腫組HIF-1αmRNA、Bax mRNA、Bax/Bcl-2水平較正常對(duì)照組、IH組、肺氣腫組升高(均P0.05),Bcl-2 mRNA水平較正常對(duì)照組、肺氣腫組降低(均P0.05),較IH組無顯著差異(P0.05);HIF-1α與Bax、Bax/Bcl-2呈正相關(guān)(r分別為0.732、0.699),與Bcl-2呈負(fù)相關(guān)(r=-0.705)。結(jié)論 IH合并肺氣腫可誘導(dǎo)肝細(xì)胞HIF-1αmRNA、Bax mRNA、Bax/Bcl-2表達(dá)上調(diào),HIF-1α與Bax、Bcl-2相關(guān),HIF-1α可能通過調(diào)控凋亡基因Bax、Bcl-2的表達(dá)來參與肝細(xì)胞的凋亡。
[Abstract]:Objective to investigate the effect of intermittent hypoxia (IH) combined with emphysema on the expression of Bcl-2 in rat liver hypoxia inducible factor-1 偽 (HIF-1 偽) and the role of HIF-1 偽 in hepatocyte apoptosis.Every day at 8:00 and 18:00 in the fumigation box 30 min each of IH combined with emphysema group, the combination of the above mentioned IH group and emphysema group.After 14 weeks of exposure, the rats were killed and the expression of HIF-1 偽 Baxon Bcl-2 in liver tissue was measured by real-time fluorescence quantitative RT-PCR.Results HIF-1 偽 mRNA-Bax mRNA-Bax mRNA-Bax mRNA-Bax-Bcl-2 in IH with emphysema group was higher than that in normal control group.Conclusion IH combined with emphysema can induce the expression of HIF-1 偽 mRNA-Bax mRNA-Bax mRNA-Bax / Bcl-2 in hepatocytes and up-regulate the expression of Bax mRNA-Bax mRNA-Bax mRNA-Bax / Bcl-2 in hepatocytes.
