BQ123對(duì)大鼠肢體缺血-再灌注后肺損傷的影響

【摘要】:目的觀(guān)察BQ123對(duì)大鼠肢體缺血-再灌注(I-R)后肺損傷的影響。方法雄性SD大鼠30只,隨機(jī)分為對(duì)照組、I-R組和BQ123組,各10只。于缺血4 h再灌注4 h后,采用放射免疫法測(cè)定肺組織中內(nèi)皮素(ET)-1的含量,用生物化學(xué)法測(cè)定髓過(guò)氧化物酶(MPO)活性和丙二醛(MDA)含量,采用酶聯(lián)免疫吸附法檢測(cè)P-選擇素的濃度,免疫組織化學(xué)方法檢測(cè)肺組織中凋亡相關(guān)因子Fas、Bcl-2和凋亡蛋白酶(caspase-3)的表達(dá)情況,利用原位末端標(biāo)記法檢測(cè)肺細(xì)胞凋亡情況。結(jié)果與對(duì)照組比較,I-R組肺組織ET-1、MPO、MDA和P-選擇素均明顯增加。肺組織Fas、Bcl-2和Caspase-3蛋白表達(dá)明顯上調(diào),其吸光度值分別為0.294±0.003,0.108±0.005,0.174±0.003,凋亡細(xì)胞百分率明顯增高[(18.83±2.86)%](P0.01);與I-R組比較,BQ123組ET-1、MPO、MDA、P-選擇素含量明顯下降。Bcl-2表達(dá)明顯升高(0.128±0.005),Fas和Caspase-3的表達(dá)明顯減弱,分別為0.115±0.007,0.159±0.006,細(xì)胞凋亡率下降[(10.67±2.16)%](P0.01)。結(jié)論 BQ123可能通過(guò)抑制肺細(xì)胞凋亡相關(guān)蛋白的表達(dá)對(duì)肢體I-R后肺損傷產(chǎn)生保護(hù)作用。
[Abstract]:Objective To observe the effects of BQ123 on ischemia - reperfusion in rats (I-R) after lung injury. Methods 30 male SD rats were randomly divided into control group, I-R group and BQ123 group, 10 rats in each. In the ischemia 4 h reperfusion after 4 h were determined by radioimmunoassay in lung tissue endothelin (ET) -1 content, determination of myeloperoxidase by biochemical method (MPO) activity and malondialdehyde (MDA) content were detected by enzyme-linked immunosorbent assay P- selectin concentration, apoptosis related factors in lung tissue were detected by immunohistochemical method in Fas, Bcl-2 and apoptosis protease (caspase-3) expression and detection of lung the cell apoptosis by TUNEL method. Results compared with the control group, I-R group, ET-1 in lung tissue, MPO, MDA and P- selectin were significantly increased.
