【摘要】：目的：通過彈性蛋白主動(dòng)免疫及熏煙制造豚鼠肺氣腫模型，觀察各模型中肺組織和心肌組織基質(zhì)金屬蛋白酶9,12（MMP-9、MMP-12）及干擾素介導(dǎo)蛋白10（IP-10）的表達(dá)情況及組織破壞情況，探究MMP-9、MMP-12及IP-10在自體免疫反應(yīng)所致肺氣腫形成過程中的作用。 方法：將24只健康成年的雄性豚鼠隨機(jī)分為3組：主動(dòng)免疫組、熏煙組、對(duì)照組。主動(dòng)免疫組采用皮下多點(diǎn)注射人彈性蛋白與佐劑的混懸液；熏煙組采用傳統(tǒng)熏煙造模；對(duì)照組不做任何處理，，共飼養(yǎng)12周。造模滿12周后各組動(dòng)物肺臟切片行HE染色，觀察肺臟組織形態(tài)，測量肺泡半徑。進(jìn)行肺組織及心肌組織的病理形態(tài)觀察。并用免疫組織化學(xué)法觀察各組肺組織及心肌組織中的MMP-9、MMP-12及IP-10蛋白的表達(dá)情況。數(shù)據(jù)進(jìn)行統(tǒng)計(jì)學(xué)分析。 結(jié)果： 1.肺臟組織病理形態(tài)觀察顯示：熏煙組和主動(dòng)免疫組豚鼠肺臟均出現(xiàn)肺泡結(jié)果的破壞，肺泡腔的增大，符合肺氣腫的病理形態(tài)改變。熏煙組和主動(dòng)免疫組的豚鼠肺泡半徑均較對(duì)照組明顯增大（P 0.05和P 0.05），平均肺泡間隔厚度較對(duì)照組比較有明顯差異（P 0.05和P 0.05）。 2.主動(dòng)免疫組及熏煙組肺組織及心肌組織中MMP-9、MMP-12及IP-10蛋白的表達(dá)均較對(duì)照組增多(P 0.05)。 3.主動(dòng)免疫組肺組織中MMP-9、MMP-12的蛋白表達(dá)與熏煙組比較無明顯差別(P＞0.05)。主動(dòng)免疫組肺組織中的IP-10蛋白的相對(duì)表達(dá)較熏煙組有明顯的增多，且有明顯的統(tǒng)計(jì)學(xué)差異(P 0.05)。 4.主動(dòng)免疫組心肌組織中MMP-9、MMP-12及IP-10蛋白的相對(duì)表達(dá)較熏煙組明顯增多(P0.05)。 結(jié)論： 1.熏煙組豚鼠體內(nèi)可能存在抗肺組織的自身抗體，彈性蛋白抗體自體免疫可能參與誘發(fā)豚鼠肺氣腫的形成。 2.MMP-9、MMP-12及IP-10在自體免疫反應(yīng)所致肺氣腫的發(fā)生發(fā)展中有重要的促進(jìn)作用。
[Abstract]:Objective: through the active immunization of elastin and smoked guinea pig lung tissue and pulmonary emphysema model, observation of myocardial tissue in each model of matrix metalloproteinase 9,12 (MMP-9, MMP-12) and interferon mediated protein 10 (IP-10) expression and tissue damage, explore MMP-9, the role of MMP-12 and IP-10 in the formation of autoimmune the reaction induced emphysema.
Methods: 24 healthy adult male guinea pigs were randomly divided into 3 groups: active immunization group, smoking group and control group. Active immunization group using suspension injected human elastin and adjuvant; fumigation group using traditional fumigation model; the control group were fed without any treatment. The raising of 12 weeks. 12 weeks after modeling with animal lung paraffin sections were stained with HE to observe the lung tissue morphology, measurement of alveolar radius. Pathological observation of lung tissue and myocardium and lung tissues were observed and myocardial tissue MMP-9 by immunohistochemical method, the expression of MMP-12 and IP-10 protein data. Statistical analysis was performed.
