中樞BDNF對(duì)咳嗽變異性哮喘豚鼠模型的調(diào)控作用

【摘要】:目的:研究腦源性神經(jīng)營養(yǎng)因子(BDNF)在豚鼠咳嗽變異性哮喘(CVA)發(fā)生發(fā)展中的調(diào)控機(jī)制和對(duì)生長相關(guān)蛋白43(GAP-43)表達(dá)的影響。方法:選擇Hartley雄性豚鼠為研究對(duì)象,制備并鑒定CVA豚鼠模型,確定CVA發(fā)作時(shí)腦內(nèi)BDNF含量和(GAP-43)表達(dá)的變化。人為改變CVA豚鼠中樞BDNF含量,觀察外周氣道炎癥和中樞GAP-43表達(dá)的相應(yīng)改變。結(jié)果:制備的CVA豚鼠模型通過鑒定符合要求。(1)模型組豚鼠中樞BDNF和GAP-43表達(dá)量明顯高于生理鹽水對(duì)照組(P0.05)。(2)與中樞注射人工腦脊液的CVA豚鼠相比,注射外源性BDNF可以增加肺組織氣道周圍的物質(zhì)P免疫陽性反應(yīng)物并加劇外周氣道炎癥,同時(shí)中樞GAP-43免疫陽性反應(yīng)物分布增多(154.3±25.7 vs 78.2±22.8,P0.01)。結(jié)論:中樞BDNF在豚鼠CVA發(fā)生發(fā)展中具有調(diào)控作用,這種作用可能通過調(diào)節(jié)GAP-43表達(dá)產(chǎn)生。
[Abstract]:Objective: to study the regulatory mechanism of brain-derived neurotrophic factor (BDNF) in the pathogenesis and development of guinea pig cough variant asthma (CVA) and its effect on the expression of growth-associated protein 43 (GAP-43). Methods: male guinea pigs with Hartley were selected as subjects. To prepare and identify the CVA guinea pig model and determine the changes of BDNF content and GAP-43 expression in the brain during the attack of CVA. Results: the expression of BDNF and GAP-43 in guinea pigs in the model group was significantly higher than that in the normal saline control group (P 0.05) and central injection. Results: the expression of BDNF and GAP-43 in the guinea pig model was higher than that in the normal saline control group (P 0.05) and the expression of GAP-43 was higher than that in the normal saline control group (P 0.05). Compared with CVA guinea pigs with artificial cerebrospinal fluid, Injection of exogenous BDNF can increase the substance P immunoreactive substance around the airway of lung tissue and aggravate the peripheral airway inflammation. At the same time, the distribution of GAP-43 immunoreactive substances in central nervous system increased 154.3 鹵25.7 vs 78.2 鹵22.8P0.01.Conclusion: central BDNF plays a regulatory role in the pathogenesis and development of CVA in guinea pigs, which may be produced by regulating the expression of GAP-43.
