低頻超聲增強(qiáng)萬(wàn)古霉素對(duì)導(dǎo)管表皮葡萄球菌生物膜影響的體內(nèi)實(shí)驗(yàn)

【摘要】:【目的】建立兔導(dǎo)管相關(guān)性表皮葡萄球菌(S.epidermidis)生物膜(biofilm,BF)感染模型,探討體內(nèi)低頻超聲聯(lián)合萬(wàn)古霉素對(duì)表皮葡萄球菌BF的干預(yù)作用。 【方法】建立表皮葡萄球菌BF體外模型,將導(dǎo)管植入兔皮下,建立兔表皮葡萄球菌導(dǎo)管BF感染模型,分為空白對(duì)照組,單獨(dú)低頻超聲組,單獨(dú)萬(wàn)古霉素組,,低頻超聲聯(lián)合萬(wàn)古霉素組。涂板菌落形成單位(CFU)計(jì)數(shù)法計(jì)算導(dǎo)管細(xì)菌數(shù);導(dǎo)管表皮葡萄球菌BF結(jié)構(gòu)的改變通過(guò)掃描電鏡(SEM)來(lái)觀察;皮膚、心臟、肝臟及腎臟病理組織切片檢查炎癥反應(yīng)。 【結(jié)果】成功建立導(dǎo)管表皮葡萄球菌感染的兔模型。各組導(dǎo)管log10(CFU/ml)計(jì)數(shù)結(jié)果分別:空白對(duì)照組(6.157±0.046)/ml,單獨(dú)低頻超聲組(6.165±0.072)/ml,單獨(dú)萬(wàn)古霉素組(5.195±0.047)/ml,低頻超聲聯(lián)合萬(wàn)古霉素組(4.842±0.058)/ml。說(shuō)明單獨(dú)低頻超聲對(duì)BF內(nèi)細(xì)菌沒(méi)有殺滅作用,單獨(dú)萬(wàn)古霉素能有效地殺滅BF內(nèi)細(xì)菌,與空白對(duì)照組相比CFU減少,p<0.01;低頻超聲聯(lián)合萬(wàn)古霉素組效果更加明顯。SEM結(jié)果顯示低頻超聲對(duì)BF結(jié)構(gòu)無(wú)影響;與各單獨(dú)作用組相比,低頻超聲聯(lián)合萬(wàn)古霉素能明顯地破壞BF,使其結(jié)構(gòu)變得更加疏松。皮膚病理組織切片顯示低頻超聲聯(lián)合萬(wàn)古霉素組,皮下隧道炎癥明顯減輕?瞻讓(duì)照組、單獨(dú)低頻超聲組、單獨(dú)萬(wàn)古霉素組,低頻超聲聯(lián)合萬(wàn)古霉素組心臟、肝臟及腎臟組織未見(jiàn)異常。 【結(jié)論】低頻超聲無(wú)法破壞表皮葡萄球菌BF結(jié)構(gòu),萬(wàn)古霉素對(duì)表皮葡萄球菌BF結(jié)構(gòu)有一定的破壞。低頻超聲聯(lián)合萬(wàn)古霉素能夠更顯著的清除導(dǎo)管表皮葡萄球菌BF結(jié)構(gòu),說(shuō)明低頻超聲能夠增強(qiáng)萬(wàn)古霉素對(duì)BF結(jié)構(gòu)的破壞及細(xì)菌的殺滅能力。
[Abstract]:[objective] to establish a rabbit model of catheter-associated staphylococcus epidermidis biofilm (BFF) infection and to investigate the effect of low frequency ultrasound combined with vancomycin on staphylococcus epidermidis BF in vivo.[methods] the model of staphylococcus epidermidis BF in vitro was established, and the infective model of staphylococcus epidermidis was established by implanting the catheter under rabbit skin. The model was divided into blank control group, low frequency ultrasound group and vancomycin group.Low frequency ultrasound combined with vancomycin group.The bacterial count of catheterization was calculated by CFU method, the changes of BF structure of Staphylococcus epidermidis were observed by scanning electron microscope (SEM), and the inflammatory reaction was detected by pathological sections of skin, heart, liver and kidney.[results] the rabbit model of staphylococcus epidermidis infection was established successfully.The count results of CFU / ml in each group were as follows: the blank control group (6.157 鹵0.046) / ml, the low frequency ultrasonic group (6.165 鹵0.072) / ml, the vancomycin group (5.195 鹵0.047) / ml, the low frequency ultrasound combined with vancomycin group (4.842 鹵0.058) / ml respectively.The results showed that low frequency ultrasound alone did not kill bacteria in BF, and vancomycin alone could effectively kill bacteria in BF.Compared with the blank control group, CFU decreased P < 0.01, the effect of low frequency ultrasound combined with vancomycin was more obvious. The results showed that low frequency ultrasound had no effect on BF structure.Low frequency ultrasound combined with vancomycin can significantly destroy BFs and make their structures more loose.The pathological sections of skin showed that the subcutaneous tunnel inflammation was alleviated obviously in low frequency ultrasound combined with vancomycin group.There were no abnormalities in heart, liver and kidney in blank control group, single low frequency ultrasound group, single vancomycin group and low frequency ultrasound combined vancomycin group.[conclusion] low frequency ultrasound can not destroy the BF structure of Staphylococcus epidermidis, and vancomycin can destroy the BF structure of Staphylococcus epidermidis.Low frequency ultrasound combined with vancomycin could significantly remove staphylococcus epidermidis BF structure, indicating that low frequency ultrasound could enhance vancomycin's ability to destroy BF structure and kill bacteria.
