【摘要】：試驗(yàn)旨在評(píng)價(jià)當(dāng)歸補(bǔ)血湯提取物安全性,為臨床安全用藥提供試驗(yàn)依據(jù)。選取120只SD大鼠(雌雄各半)隨機(jī)分為高、中、低劑量組和空白對(duì)照組,藥物組分別按60、30、15 g/kg體重劑量灌胃給藥,空白對(duì)照組給予等體積蒸餾水,連續(xù)給藥28 d,停藥后觀察14 d。結(jié)果顯示,給藥結(jié)束時(shí),高劑量組雄性大鼠出現(xiàn)體重增長(zhǎng)減緩(P0.01)和飼料消耗量降低(P0.01),雄性大鼠白細(xì)胞數(shù)(WBC)、淋巴細(xì)胞(Lym)均顯著升高(P0.05),丙氨酸氨基轉(zhuǎn)移酶(ALT)、總膽固醇(TC)、葡萄糖(GLU)均極顯著降低(P0.01),血小板(PLT)顯著減少(P0.05);雌性大鼠紅細(xì)胞(RBC)、血紅蛋白(HGB)、ALT均極顯著減少(P0.01)。中劑量組大鼠ALT和堿性磷酸酶(ALP)顯著或極顯著降低(P0.05;P0.01)。停藥第14天雄性大鼠網(wǎng)織紅細(xì)胞(RET)顯著增多(P0.05),雌性大鼠HGB顯著減少(P0.05)。停藥后可恢復(fù)正常,其余相關(guān)檢測(cè)指標(biāo)均未見(jiàn)異常。各臟器組織病理學(xué)檢查均未見(jiàn)異常。結(jié)果表明當(dāng)歸補(bǔ)血湯提取物毒性低,可以安全用于動(dòng)物。
[Abstract]:The purpose of the experiment was to evaluate the safety of Danggui Buxue decoction extract and to provide experimental basis for clinical safety drug use.120 Sprague-Dawley rats (half male and female) were randomly divided into three groups: high, middle, low dose and blank control group. The drug group was given intragastrically at a body weight dose of 60 ~ 30g / 15 g/kg. The blank control group was given distilled water of the same volume for 28 days and observed for 14 days after withdrawal.The results showed that, at the end of the administration,In the high dose group, the weight gain of male rats decreased P0.01) and feed consumption decreased P0.01a. The white blood cell count of male rats increased significantly (P0.05), and the alanine aminotransferase (alt), total cholesterol (TCU) and glucose (GLU) decreased significantly.P0. 01, PLT) significantly decreased P0. 05%, while in female rats, RBC, HGBM and alt were significantly decreased in P0. 01%, and in female rats, P0. 01%, P 0. 01, P 0. 01, P 0. 01, P 0. 01, P 0. 01, respectively.In the middle dose group, ALT and alkaline phosphatase (ALP) decreased significantly or extremely significantly.On the 14th day after withdrawal, the reticulocyte (RET) of male rats increased significantly, while the HGB of female rats decreased significantly.After withdrawal, the drug can return to normal, and the other related indicators are not abnormal.No abnormality was found in histopathological examination of all organs.The results showed that the extract of Danggui Buxue decoction had low toxicity and could be safely used in animals.
