SARS-CoV-2受體ACE2的單細(xì)胞RNA表達(dá)譜在小鼠呼吸道中的表達(dá)分布

目的了解SARS-CoV-2受體血管緊張素轉(zhuǎn)化酶2(angiotensin-converting enzyme 2,ACE2)在小鼠呼吸道及口腔(包括舌頭和頰黏膜上皮)的分布。方法利用單細(xì)胞公共數(shù)據(jù)庫(kù)和最新的單細(xì)胞RNASeq技術(shù),對(duì)小鼠氣管,肺臟進(jìn)行單細(xì)胞數(shù)據(jù)可視化分析,對(duì)頰黏膜上皮進(jìn)行重新聚類(lèi)分析。結(jié)果小鼠肺臟少量II型肺泡上皮細(xì)胞(AT2)表達(dá)ACE2,與人體相似,氣管中各類(lèi)型細(xì)胞散在表達(dá)ACE2,在小鼠舌頭的角質(zhì)形成細(xì)胞(41.74%)表達(dá)ACE2,在小鼠的分化期口腔頰黏膜上皮細(xì)胞中表達(dá)ACE2,并且與肺臟ACE2+ AT2相同,也表達(dá)病毒進(jìn)程相關(guān)基因,部分基因呈現(xiàn)特異性表達(dá)。結(jié)論冠狀病毒可能定植于口腔中,并通過(guò)舌頭黏膜等口腔器官分泌物進(jìn)行傳播。
