SSTR4,SSTR5在隱孢子蟲感染后腸易激綜合征大鼠空腸表達(dá)研究

目的:研究隱孢子蟲感染SD大鼠乳鼠后,空腸生長抑素受體(Somatostatin receptor,SSTR)4和SSTR5表達(dá)水平的變化,及奧曲肽對(duì)感染后腸易激綜合征(Post-infectious irritablr bowel syndrome,PI-IBS)大鼠空腸 SSTR4 和 SSTR5 表達(dá)的影響。方法:對(duì)贈(zèng)與得到的隱孢子蟲卵囊進(jìn)行鑒定和計(jì)數(shù),對(duì)SD大鼠進(jìn)行免疫抑制后灌胃給予106個(gè)隱孢子蟲卵囊,收集感染后第3-15天糞便,對(duì)隱孢子蟲卵囊進(jìn)行純化,計(jì)數(shù)后將濃度稀釋至109·L-1。將128只5日齡SD大鼠乳鼠分為空白對(duì)照組、給藥對(duì)照組、感染組和給藥組,另外10只為確認(rèn)感染組(5只感染隱孢子蟲,5只作為正常對(duì)照組)。分組后感染組和給藥組64只的5日齡SD大鼠乳鼠灌胃給予隱孢子蟲卵囊(109·L-1,每只0.1 mL),空白對(duì)照組和給藥對(duì)照組64只的5日齡SD大鼠乳鼠灌胃給予PBS(0.1ml)。隱孢子蟲灌胃第二天記為感染后第1天。感染后第7天,處死確認(rèn)感染組大鼠乳鼠,取腸道各段,固定,HE染色,主要觀察空腸是否寄生隱孢子蟲,確認(rèn)是否5只全部感染隱孢子蟲。給藥對(duì)照組和給藥組...
