檢測(cè)基因及樣本類(lèi)型對(duì)人感染H7N9禽流感病毒實(shí)驗(yàn)室檢測(cè)結(jié)果的影響
[Abstract]:Objective to detect the H7N9 influenza virus related genes in the samples of H7N9 avian influenza virus infection cases and close contacts in Beijing, and to analyze the effect of different target genes and different types of samples on the detection results. Methods from April to July 2013, samples of throat swabs, sputum, trachea lavage fluid and serum from Beijing people infected with H7N9 avian influenza and close contacts were collected, and virus RNA, was extracted to detect the (influenza A matrix protein,Am gene of influenza A virus matrix protein. Hemagglutinin 7 (hemagglutinin 7 H7) gene and neuraminidase 9 (neuraminidase 9 N9) gene were analyzed statistically. Results the positive rates of Am gene, H7 gene and N9 gene were 68.3% and 41.5% respectively. In quantitative analysis, the quantitative limits of Am gene, H7 gene and N9 gene were 10 copies/ml,10copies/ml and 100 copies/ml., respectively. The detection rate of viral nucleic acid in trachea lavage fluid samples was higher than that in serum and conjunctival secretions. Conclusion in the detection of human infection with H7N9 avian influenza, the lower respiratory tract samples such as trachea lavage fluid should be used as far as possible, and the test results should be judged synthetically to provide reference for clinical treatment.
