【摘要】：目的探討HBV感染對(duì)肝臟自然殺傷細(xì)胞(NK細(xì)胞)和固有淋巴樣細(xì)胞(ILC)22的影響,為闡明HBV感染誘導(dǎo)機(jī)體固有免疫應(yīng)答的機(jī)制提供理論和實(shí)驗(yàn)依據(jù)。方法選取6~8周齡雄性BALB/c小鼠10只,隨機(jī)分為實(shí)驗(yàn)組和對(duì)照組,每組各5只。實(shí)驗(yàn)組小鼠經(jīng)尾靜脈高壓注射含10μg HBV全基因組質(zhì)粒的生理鹽水(體積相當(dāng)于9%小鼠體質(zhì)量);對(duì)照組小鼠僅注射生理鹽水。4 d后處死小鼠,分離肝臟內(nèi)淋巴細(xì)胞,應(yīng)用流式細(xì)胞術(shù)檢測(cè)肝臟內(nèi)淋巴細(xì)胞中NK細(xì)胞亞群和ILC22細(xì)胞亞群的比例。組間比較采用t檢驗(yàn)。結(jié)果高壓尾靜脈注射HBV全基因組質(zhì)�？烧T導(dǎo)小鼠產(chǎn)生高水平的HBs Ag和HBe Ag,伴有血清ALT水平升高。實(shí)驗(yàn)組小鼠肝臟內(nèi)NK細(xì)胞的比例較對(duì)照組小鼠顯著升高[(25.90±4.92)%vs(12.98±2.13)%,t=3.811,P=0.003],但CD127+NK細(xì)胞和CD127-NK細(xì)胞亞群在NK細(xì)胞中所占比例在2組之間差異均無(wú)統(tǒng)計(jì)學(xué)意義(P值均0.05)。實(shí)驗(yàn)組小鼠肝臟內(nèi)NKp46+ILC22細(xì)胞亞群比例較對(duì)照組明顯升高[(36.05±6.85)%vs(10.22±3.54)%,t=7.372,P0.001],但NKp46-ILC22細(xì)胞亞群的比例在2組之間差異無(wú)統(tǒng)計(jì)學(xué)意義(P0.05)。結(jié)論 HBV感染可誘導(dǎo)肝臟內(nèi)NK細(xì)胞和NKp46+ILC22水平升高,從而促進(jìn)肝臟固有免疫應(yīng)答。
[Abstract]:Objective to investigate the effects of HBV infection on natural killer cells (NK cells) and lymphoid like cells (ILC) 22) of the liver, and to provide theoretical and experimental evidence for elucidating the mechanism of the innate immune response induced by HBV infection. Methods Ten male BALB/c mice aged 6 and 8 weeks were randomly divided into experimental group and control group with 5 mice in each group. The mice in the experimental group were injected with saline containing 10 渭 g HBV genome plasmid through caudal vein hyperbaric injection (volume equivalent to 9% of the body mass of mice), and the control group mice were killed after injection of saline only for 4 days, and the lymphocytes in the liver were isolated. The proportion of NK cell subsets and ILC22 cell subsets in liver lymphocytes was detected by flow cytometry. T test was used for comparison between groups. Results High pressure tail vein injection of HBV genome plasmid could induce high levels of HBs Ag and HBe Ag, with elevated serum ALT level in mice. The percentage of NK cells in the liver of the experimental group was significantly higher than that of the control group [(25.90 鹵4.92) vs (12.98 鹵2.13)], but the proportion of CD127 NK cells and CD127-NK subsets in the NK cells was not significantly different between the two groups (P < 0. 05). The percentage of NKp46 ILC22 cell subsets in the liver of the experimental group was significantly higher than that of the control group [(36.05 鹵6.85) vs (10.22 鹵3.54)], but there was no significant difference in the proportion of NKp46-ILC22 cell subsets between the two groups (P0.05). Conclusion HBV infection can induce the increase of NK cells and NKp46 ILC22 levels in the liver and promote the inherent immune response of the liver.
