【摘要】：日本血吸蟲(chóng)病的篩查和診斷在血吸蟲(chóng)病的防治中處于重要的地位,而核酸診斷是其潛在的優(yōu)良檢測(cè)方法。擬建立一套完整的"可視化日本血吸蟲(chóng)核酸恒溫?cái)U(kuò)增檢測(cè)試劑",用于基層血防單位對(duì)日本血吸蟲(chóng)病的篩查和診斷等工作。用腹壁拔毛貼片法給新西蘭大白兔感染日本血吸蟲(chóng),用多種核酸提取方法抽提其血清或血漿中的基因組DNA,并用"可視化日本血吸蟲(chóng)核酸恒溫?cái)U(kuò)增檢測(cè)試劑"對(duì)其進(jìn)行檢測(cè)。結(jié)果表明,該研究所設(shè)計(jì)的檢測(cè)方法可以方便地檢測(cè)到100條日本血吸蟲(chóng)尾蚴感染的新西蘭大白兔,說(shuō)明可視化核酸恒溫?cái)U(kuò)增技術(shù)在日本血吸蟲(chóng)病的篩查和診斷中具有潛在的應(yīng)用可能性。
[Abstract]:The screening and diagnosis of schistosomiasis japonicum plays an important role in the prevention and control of schistosomiasis, and nucleic acid diagnosis is a potential good detection method. A complete set of "visualized nucleic acid constant temperature amplification reagent" for screening and diagnosis of schistosomiasis japonicum was established. New Zealand white rabbits were infected with Schistosoma japonicum by abdominal wall plucking patch method. The genomic DNA in serum or plasma was extracted by a variety of nucleic acid extraction methods, and the genomic DNA was detected by "visualized Schistosoma japonicum nucleic acid constant temperature amplification reagent". The results showed that 100 New Zealand white rabbits infected with Schistosoma japonicum cercariae could be easily detected by the method designed by the Institute. The results indicate that the visualization nucleic acid isothermal amplification technique has potential application in the screening and diagnosis of schistosomiasis japonicum.
