【摘要】：目的探討樹突狀細(xì)胞相關(guān)C型凝集素-1(Dectin-1)在熱處理光滑假絲酵母菌刺激大鼠氣道上皮細(xì)胞(RTECs)中的作用及對IL-10和TNF-α表達(dá)的影響。方法將體外培養(yǎng)的RTECs隨機(jī)分為3組:對照組(RTECs+無菌生理鹽水)、真菌刺激組(RTECs+熱處理光滑假絲酵母菌)、抑制劑干預(yù)組(RTECs+昆布多糖+熱處理光滑假絲酵母菌),分別孵育0、2、4、6 h后終止,MTT法檢測細(xì)胞存活率,Western Blot法檢測Dectin-1表達(dá),ELISA法檢測IL-10和TNF-α的表達(dá)。結(jié)果熱處理光滑假絲酵母菌破壞細(xì)胞結(jié)構(gòu),降低細(xì)胞存活率。在培養(yǎng)開始時(0h),3組RTECs細(xì)胞存活率以及Dectin-1、IL-10、TNF-α表達(dá)水平比較,差異均無統(tǒng)計學(xué)意義(均P0.05)。培養(yǎng)2、4、6 h后,真菌刺激組、抑制劑干預(yù)組Dectin-1、IL-10、TNF-α表達(dá)水平均高于對照組,抑制劑干預(yù)組Dectin-1、IL-10、TNF-α表達(dá)水平均低于真菌刺激組(均P0.05)。除抑制劑干預(yù)組0h與2 h IL-10表達(dá)量比較,差異無統(tǒng)計學(xué)意義之外,真菌刺激組和抑制劑干預(yù)組組內(nèi)不同時間段Dectin-1、IL-10、TNF-α表達(dá)水平比較,差異均有統(tǒng)計學(xué)意義(均P0.05)。結(jié)論 Dectin-1是RTECs識別熱處理光滑假絲酵母菌的重要受體,并誘導(dǎo)其釋放IL-10和TNF-α,介導(dǎo)炎癥反應(yīng)的發(fā)生。
[Abstract]:Objective to investigate the role of dendritic cell-related C-type lectin-1 (Dectin-1) in heat treated rat airway epithelial cells stimulated by Candida smoothing and its effect on the expression of IL-10 and TNF- 偽. Methods RTECs cultured in vitro were randomly divided into three groups: control group (control group), fungal stimulation group (RTECs) heat treated Candida cerevisiae, and inhibitor intervention group (control group), which were incubated respectively by heat treated Candida cerevisiae. The expression of Dectin-1 was detected by Western Blot assay and the expression of IL-10 and TNF- 偽 was detected by Elisa. Results the cell structure was destroyed and the cell survival rate was decreased after heat treatment. At the beginning of culture, there was no significant difference in the survival rate of RTECs cells and the expression of IL-10 TNF- 偽 in Dectin-1 (all P 0.05). The expression of TNF- 偽 in Dectin-1 and inhibitor intervention group was higher than that in control group, and the expression level of Dectin-1 IL-10 TNF- 偽 in inhibitor intervention group was lower than that in fungal stimulation group (all P 0.05). The expression of Dectin-1 IL-10 TNF- 偽 in the fungal stimulation group and the inhibitor intervention group was significantly higher than that in the control group at 0 h and 2 h (all P < 0.05), except that the expression of Dectin-1 IL-10 TNF- 偽 was significantly different between the two groups at 0 h and 2 h (P < 0.05). Conclusion Dectin-1 is an important receptor for RTECs to recognize the heat treated Candida smoothing, and induce the release of IL-10 and TNF- 偽, and mediate the inflammatory reaction.
