【摘要】：目的:探索分枝桿菌噬菌體Guo1對(duì)結(jié)核休眠菌的復(fù)蘇作用。方法:采用缺鉀Sauton培養(yǎng)基培養(yǎng)對(duì)數(shù)生長(zhǎng)期結(jié)核菌構(gòu)建結(jié)核休眠菌模型;采用藥敏檢測(cè)、透射電子顯微鏡(電鏡)觀察和金胺O-尼羅紅雙熒光染色作為模型檢測(cè)方法;采用菌落計(jì)數(shù)作為復(fù)蘇的檢測(cè)指標(biāo)。結(jié)果:缺鉀結(jié)核休眠菌模型構(gòu)建成功�；旌吓囵B(yǎng)第1天時(shí),空白對(duì)照組、上清組、TM4組、Guo1組菌落計(jì)數(shù)分別為(2.67±0.58)×10~2、(3.00±1.00)×10~2、(2.67±0.58)×10~2、(2.33±0.58)×10~2CFU/m L;培養(yǎng)第15天時(shí),各組菌落計(jì)數(shù)分別為(7.33±1.53)×10~4、(6.67±2.08)×10~(10)、(3.00±1.00)×10~(10)、(3.33±1.53)×10~(10)CFU/m L。結(jié)論:分枝桿菌噬菌體Guo1能夠復(fù)蘇結(jié)核休眠菌。
[Abstract]:Objective: to explore the resuscitation effect of Mycobacterium bacteriophage Guo1 on Mycobacterium tuberculosis dormancy bacteria. Methods: the mycobacterium tuberculosis dormancy bacteria model was established by using potassium deficiency Sauton culture medium culture logarithmic growth phase tuberculous bacilli, drug sensitivity test, transmission electron microscope (TEM) observation and gold amine O- Nile red double fluorescence staining as the model detection method. Colony count was used as the index of resuscitation. Results: the model of dormancy bacteria was successfully constructed.
