抗OPN抗體對沙鼠肝多房棘球蚴組織的侵襲性影響的遠期觀察結果及意義

【摘要】:目的:在沙鼠肝多房棘球蚴動物模型基礎上,在給予抗OPN抗體干預沙鼠多房棘球蚴后,通過檢測MMP-2、TGF-β1因子的表達,探究抗OPN抗體與多房棘球蚴泡囊浸潤性生長、轉(zhuǎn)移可能存在的相關性。 方法:我們使用多房棘球蚴原頭節(jié)混懸液(0.15ml/只),在開腹直視下對沙鼠肝臟穿刺接種,制造肝多房棘球蚴動物模型。實驗中取4-6周齡健康雌性長爪沙鼠120只(305)g,隨機分為:模型組、實驗組、對照組。模型組僅接種多房棘球蚴組,實驗組接種多房棘球蚴同時外源性注射抗OPN抗體干預,而對照組在接種多房棘球蚴同時予以注射兔血清(抗OPN抗體溶劑)干預。再將每組肝多房棘球蚴沙鼠隨機分成6小組,每小組10只,為感染140d、180d、220d、260d組。于相應時間點處死各組沙鼠,觀察多房棘球蚴的生長、轉(zhuǎn)移等情況;免疫組化法測定肝多房棘球蚴組織中MMP-2、TGF-β1因子的表達。 結果: 1.140天、180天、220天時,實驗組與對照組、實驗組與模型組囊濕重有統(tǒng)計學差異(F值分別為8.647,36.96,9.12,p<0.05),而對照組與模型之間比較無統(tǒng)計學差異(P㧐0.05)。 2.觀察泡狀棘球蚴轉(zhuǎn)移現(xiàn)象,胸廓淋巴結轉(zhuǎn)移率較高,而肺轉(zhuǎn)移少見,未見腦轉(zhuǎn)移灶。統(tǒng)計結果顯示:140天、180天、組時實驗組與對照組、實驗組與模型組胸廓淋巴結轉(zhuǎn)移率有顯著性差異(P㩳0.05),220天組時實驗組與對照組,實驗組與模型組胸廓淋巴結轉(zhuǎn)移率無顯著性差異(P㧐0.05)。 3. HE染色結果顯示:在140天、180天、220天時間段,實驗組與對照組、實驗組與模型組間E.m囊壁病理的改變無顯著性差異(P㧐0.05)。 4.免疫組化染色結果顯示:肝多房棘球蚴組織中可見MMP-2、TGF-β1因子不同程度的表達,定位于細胞漿,主要分布在肝多房棘球蚴纖維囊壁的生發(fā)層,細胞漿中可見黃色、棕黃色或棕褐色等陽性顆粒;MMP-2在實驗組、對照組與模型組免疫組化染色陽性細胞率比較差異無統(tǒng)計學意義(P㧐0.05)。而實驗組TGF-β1陽性細胞率在在20天、60天、100天、140天、180天、及220天時間段,,TGF-β1在實驗組與對照組、實驗組與模型組間免疫組化染色陽性細胞率比較無顯著性差異(P0.05),100天組、140天組、180天組時TGF-β1的陽性細胞表率達在實驗組與對照組、實驗組與模型組比較有顯著性差異(P㩳0.05)。 結論:MMP-2、TGF-β1因子共同參與了沙鼠肝泡狀棘球蚴多房棘球蚴侵襲轉(zhuǎn)移進程進程。通過抗OPN抗體干預可使肝多房棘球蚴組織中TGF-β1因子的表達減低,表明抗OPN抗體干預可能是抑制肝多房棘球蚴中晚期侵襲性生長的機制之一。
[Abstract]:Objective: to investigate the expression of MMP-2 TGF- 尾 1 factor in multilocularis of gerbils by anti OPN antibody, and to explore the infiltrating growth of Echinococcus multilocularis with anti OPN antibody. The possible relevance of the transfer. Methods: a multilocular echinococcus multilocularis model was established by inoculation of gerbil liver with 0.15ml / L Echinococcus multilocularis suspension. 120 healthy female gerbils aged 4-6 weeks were randomly divided into three groups: model group, experimental group and control group. The model group was only inoculated with multi-loculed echinococcus group, the experimental group was inoculated with multi-loculed echinococci and exogenous anti-OPN antibody was injected, while the control group was injected with rabbit serum (anti-OPN antibody solvent) at the same time. Each group was randomly divided into 6 groups, 10 rats in each group. The growth and metastasis of echinococcus multilocularis were observed, and the expression of MMP-2 and TGF- 尾 1 in hepatic hydatid tissues were detected by immunohistochemical method. Results: At 1.140 days after 180 days and 220 days, there was a significant difference in the wet weight of the capsule between the experimental group and the control group, the experimental group and the model group, and the F value was 8.647g / 36.96.96 / 9.12p < 0.05 respectively, but there was no significant difference between the control group and the model group (P < 0.05). 2. The metastatic rate of thoracic lymph nodes was higher than that of lung metastasis. The statistical results showed that there were significant differences between the experimental group and the control group at 140 days to 180 days, and the rates of thoracic lymph node metastasis between the experimental group and the model group at 220 days. There was no significant difference between the experimental group and the model group in the rate of thoracic lymph node metastasis. 3. The results of HE staining showed that there was no significant difference between the experimental group and the control group and between the experimental group and the model group in the pathological changes of the E m cyst wall at the time of 140 days, 180 days and 220 days, and there was no significant difference between the experimental group and the control group. 4. Immunohistochemical staining showed that the expression of MMP-2TGF- 尾 1 was localized in the cytoplasm, mainly in the germinal layer of the fibrous cyst wall of the hepatic hydatid, and yellow in the cytoplasm of the cells, and the expression of MMP-2TGF- 尾 1 was observed in different degree in the liver hydatid tissues, and the expression of MMP-2TGF- 尾 1 was localized in the cytoplasm. There was no significant difference in the positive rate of MMP-2 between the control group and the model group by immunohistochemical staining. The positive rate of TGF- 尾 1 in the experimental group was at 20 days, 60 days, 100 days, 140 days, 180 days, and 220 days, the rates of TGF- 尾 1 positive cells were found in the experimental group and the control group. There was no significant difference in the positive rate of TGF- 尾 1 between the experimental group and the model group. There was no significant difference between the experimental group and the model group. The positive cell rate of TGF- 尾 1 was higher in the experimental group and the control group than in the model group, and there was a significant difference between the experimental group and the model group. Conclusion 1: MMP-2 TGF- 尾 1 is involved in the process of invasion and metastasis of hydatid hydatid in gerbils. The expression of TGF- 尾 1 in hepatic hydatid tissues was decreased by the intervention of anti OPN antibody, which indicated that the intervention of anti OPN antibody might be one of the mechanisms of inhibiting the advanced invasive growth of hepatic hydatid.
